Background:Hyperuricemia(HUA)is a metabolic disease caused by disordered purine metabolism.The end product of purine metabolism is uric acid.If it is excreted too little and/or generated too much,the concentration of uric acid in the human serum will be too high.Hyperuricemia causes more than gouty arthritis and long-term high levels of uric acid can cause kidney damage that can cause gouty nephropathy.Jiangsuanling is Professor Hu Xuguang's experience in the addition and subtraction of the ancient formula based on the ancient formula.The effect of reducing uric acid is remarkable.However,the experimental aspects have not yet been carried out.In order to explore its specific experimental efficacy and mechanism of action,this article is intended to be validated by a mouse model of hyperuricemia.Objective: To verify the effect of Jiangsuanling on uric acid lowering in model mice with hyperuricemia and its influence on renal function,and explore its possible mechanism,so as to provide support for further development.METHODS: 84 male KM mice were randomly divided into seven groups: blank group,model group,allopurinol group,high-dose of lower acid dose group,descending acid-lowering dose group,low-dose acidlower dose group,benzbromarone group.The model of hyperuricemia was replicated by intraperitoneal injection of potassium oxonate.Seven days after the modeling,the other groups except the blank group and the model group were given the corresponding drugs according to the equivalent dose conversion table of the pharmacological experiment.The blank group and the model group were all irrigated.An equal volume of distilled water from the stomach was obtained after 14 days of administration.The biochemical test kits were used to measure the XOD activity of HUA,Cr,BUN,liver and serum.The pathological sections of kidneys were stained with inverted microscope to test the degree of renal damage.The relative expression levels of URAT1 and ABCG2 proteins were detected by immunoblotting.Fluorescent quantitative PCR was used to detect the relative expression levels of mGLUT9 and mOAT1 in the kidney.RESULTS: Jiangsuling can significantly reduce the serum uric acid value of hyperuricemia mice(P<0.01),has a significant effect of lowering uric acid,and can significantly reduce serum Cr and BUN levels(P<0.01 or P<0.05);can significantly reduce Liver tissue xanthine oxidase(XOD)and serum xanthine oxidase(XOD)activity(P<0.01 or P<0.05);significant down-regulation of URAT1 protein expression in the kidney(P<0.01),ABCG2 protein of kidney Relative expression had no significant effect;renal GLUT9 mRNA was significantly down-regulated(P<0.01 or P<0.05)and OAT1 mRNA was up-regulated(P<0.01).Conclusion: Jiangsuanling Decoction can significantly reduce the serum uric acid level in hyperuricemia mice and has the effect of preventing and treating hyperuricemia;and decoction also can reduce serum creatinine and uric acid nitrogen levels in hyperuricemia mice to protect renal function and reduce its The role of renal pathological injury.The mechanism of uric acid lowering by acid-lowering may be related to inhibiting XOD activity in liver and serum,decreasing the expression of URAT1 protein in kidneys of hyperuricemia mice,down-regulating the expression of GLUT9 mRNA in kidney,and up-regulating the expression of OAT1 mRNA in kidneys.related.
