Aloe vera is a perennial succulent evergreen herbs,belongs to the Asphodelaceae?Liliaceae?family,native to the Mediterranean and Africa,now almost all over the world were planted.Aloe variety not only,and the varieties are relatively extensive pharmacological activities,is a collection of beauty,food,health care,in a body's functional plant,with its reputation as the“plant doctor”.At present,the chemical components and pharmacological activities of aloes are correlation research reports,but merely to Aloe barbadensis Mill,for another species,Aloe arborescens Mill,there have been few reports.After the second world war,aloe was used by the Japanese to resist inflammation,radiation and burn wounds.It was considered by the Japanese as the best medicinal aloe variety.In order to explore the chemical composition of aloe and the comprehensive quality control of aloe vera,we studied the following aspects:The main studies were summarized as follows:1.Ultrasonic-assisted extraction for Aloe arborescens MillFreshly dried aloe vera leaves are dried and beaten into powder,the single-factor test combine with response surface methodology were applied in the optimization procedure of ultrasonic-assisted extraction for Aloe arborescens Mill.The best process conditions of ultrasonic-assisted was as follows:the content of methanol:80%;material-to-solvent ratio:1:15;extraction time:45min.Ultrasonic-assisted extraction was simple as well as fast,and it made sense to guide the extraction for Aloe arborescens Mill.2.Separation and Purification compounds in Aloe arborescens MillAfter fractionation by macroporous resin column chromatography,40%and 60%methanol fraction was selected based on HPLC qualitative analysis.Medium Pressure Preparative Liquid Chromatography was performed to separate aloenin,2?-?-rhamnopyranosyl-isovitexin,aloin B,aloinA,with the purity of 99.6%?98.1%?98.9%and 98.6%,respectively.The separation method established in this experiment is simple and fast and efficient,which is of great significance to the quality control and fingerprint of Aloe arborescens Mill.3.The systematical qualitative analysis of chemical components in Aloe arborescens Mill by liquid chromatography-mass spectrometry-ion trap-time-of-flightThe chemical composition of aloe powder was identified by rapid analysis.The chemical composition of aloe was analyzed by high performance liquid chromatography/electrospray-ion trap-time mass spectrometry?LCMS-IT-TOF?,and Thermo C18?250 mm×4.6 mm,5?m?column was used.The gradient elution was performed with methanol-water as the flow phase,and the flow rate was 1.0 mL·min-1;Detection wavelength of 300 nm,at the same time,adopt the positive and negative ions detection mode,the results according to the results of high resolution mass spectrometry and MS/MS fragmentation information,combined with control quality spectrum information and relevant literature,belonging to the chromatographic peak of 22,and summarizes the mass spectrum of the law of cracking.This study improves the qualitative analysis efficiency of aloe chemical composition and provides a material basis for further study.4 The content of the main components of Aloe arborescens Mill was determined by quantitative analysis multi-components by single markerThe main chemical constituents of Aloe arborescens Mill?aloenin,2?-?-rhamnopyranosyl-isovitexin,aloin B,aloin A?were established.Thermo C18?250mm×4.6 mm,5?m?chromatographic column was adopted;The gradient elution was performed with methanol-water as the flow phase,and the flow rate was 1.0 mL·min-1;The detection wavelength is 300 nm and the sample size is 10?L.Using aloinA as the internal reference,the main chemical composition of aloe by quantitative analysis multi-components by single marker was established.The method is simple and reliable.Therefore,the standard curve method can be substituted when the control product is scarce,and the quantity of various components can be accurately and quickly.This study provides a new idea for the quality evaluation of Aloe arborescens Mill and lays a solid foundation for the construction of the corresponding industry standard.5 HPLC fingerprint and chemical pattern recognition method of Aloe arborescens MillTo establish the HPLC fingerprint of Aloe arborescens Mill powder to provide reference of the effective quality control.The fingerprint of Aloe arborescens Mill powder was developed by HPLC.The separation was carried out on a Thermo-C18column?250 mm×4.6 mm,5?m?with the mobile phase consisting of methanol and water at 1.0 mL·min-1.The detection wavelength was 300nm and the sample volume was 10?L.According to the similarity evaluation,the chemometrics method was used to evaluate the quality of Aloe arborescens Mill powder.Fingerprints of 19 batches of Aloe arborescens Mill powder were established,the common mode of the fingerprint was set up with 19 common peaks,and four of them were identified.The similar degrees of 19 batches of samples were over 0.9.The samples were classified into three groups by hierarchical cluster analysis combined with principal component analysis?PCA?and orthogonal partial least squares-discriminant analysis?OPLS-DA?.The establishment of HPLC fingerprint of Aloe arborescens Mill powder and the application of chemical pattern recognition can provide a more comprehensive reference for quality control.
