Objective:1.To identify the differences of the expression of miR-125 b and PKM2 between cervical cancer SiHa cells and normal epithelium.2.To investigate the effect on the expression of PKM2 when change the expression of miR-125 b in cervical cancer SiHa cells.Methods:1.Cultivated cervical cancer SiHa cells and normal epithelial cell line(293T).The 293 T cell line was treated as negative control group(Group N).Transfected the SiHa cells,and divided it into three groups:(1)The experimental group:miR-125 b mimics(Group M)、miR-125 b inhibitor(Group I);(2)Negative control group:miR-125 b mimics negative control(Group MNC)、 mi R-125 b inhibitor negative control(Group INC);(3)Blank control group(Group B):no transfection.2.qRT-PCR technology was used to detect the espression level of miR-125 b in three groups of SiHa cells and 293 T cells.3.Immunofluorescence and Western blot technology were used to detect the expression level of PKM2 protein in three groups of SiHa cells and 293 T cells.Results:1.The expression level of miR-125 b in SiHa cells was(0.820 + 0.052)times lower than that in 293 T cells(Group N),P < 0.05.2.The results of immunofluorescence showed that the number of cells expressing PKM2 in SiHa cells(3.4 + 0.548)was higher than that in 293 T cells(0 + 0),P < 0.05.The results of Western Blot showed that the relative expression of PKM2 in the SiHa cells(0.300±0.012)was higher than that in 293 T cells(0.119 + 0.010),P < 0.05.3.After successfully transfecting SiHa cells,the results of immunofluorescence showed that the number of cells expressing PKM2 in Group M(1.0±0.707)was lower than that in Group MNC(3.6±0.548),P < 0.05;the number of cells expressing PKM2 in Group I(6.0±0.707)was higher than that in Group INC(3.6±1.342),P < 0.05.The results of Western Blot showed that the relative expression of PKM2 in Group M(0.239±0.042)was lower than that in Group MNC(0.314±0.018),P < 0.05;the relative expression of PKM2 in Group I(0.400±0.033)was higher than that in Group INC(0.340±0.042),P < 0.05.Conclusion:1.The expression of miR-125 b in cervical cancer SiHa cells is lower than that in normal epithelial.2.The expression of PKM2 in cervical cancer SiHa cells is higher than that in normal epithelial.3.Overexpression of miR-125 b may inhibit the expression of PKM2 in cervical cancer SiHa cells,which thereby inhibits the glycolysis and the growth of tumor.Knockdown of miR-125 b may increase the expression of PKM2 in cervical cancer cells,and it may promote its glycolysis and the growth of tumor..
