Objective:Cervical cancer is still a gynecologic malignancy that seriously harms the health of women worldwide.The adverse living environment,dietary factors,and cervical microbiota can all lead to cervical intraepithelial neoplasia,thereby accelerating the progression of precancerous lesions and continuously reflecting the occurrence of cervical cancer.process.The miRNA-203 is a kind of carcinogenic or study tumor-suppressor factor,and serum folic acid as the body must trace elements,DNA,RNA and protein in cells play an important role in synthetic biology,which can lead to increased sensitivity of the body of carcinogenic factors.The purpose of this study is:The expression of miRNA-203 and serum folic acid in different cervical lesions were detected.The correlation of miRNA-203 expression,folic acid and cervical intraepithelial neoplasia was analyzed.Methods:1.The patients with different cervical lesions from the Department of Obstetrics and Gynecology at the Second Hospital of Shanxi Medical University were selected as subjects.They were submitted to the cervical biopsy and diagnosed by pathology department.NC group(30 cases),LSIL group(30 cases),HSIL group(30 cases)),SCC group(30 cases).2.Access Folate Test for Detection of Serum Folic Acid.Real-time fluorescence quantitative reverse transcription PCR(real time rt-pcr)was used to detect miRNA-203expression levels in cervical tissues.3.SPSS19.0 software was used for data analysis.The comparison of quantitative data was analyzed by one-way ANOVA.The comparison between groups was performed using Dunnett-t test and LSD-T test.Spearman rank correlation and trend chi-square test were used for correlation analysis.Multivariate analysis was performed using an ordered multi-class logistic regression analysis.The test level was set to?=0.05.Results:1.The concentrations of folic acid in NC group,LSIL group,HSIL group and SCC group were 33.07±3.37nmol/L,28.44±2.76nmol/L,20.62±5.20nmol/L and 16.63±4.6nmol/L(x^-±s),The difference was statistically significant(F=98.53,P<0.01).Four groups of serum folic acid comparison,the differences were statistically significant.Among them,the serum levels of folic acid in LSIL group,HSIL group and SCC group were lower than those in NC group(P<0.001).As the degree of cervical intraepithelial neoplasia increased,the folic acid level decreased gradually.2.The expression levels of miR-203 in the NC group,LSIL group,HSIL group,and SCC group were 0.61±0.27?0.30±0.14?0.13±0.06?0.08±0.04(x^-±s)respectively.The miR-203s in the LSIL group,HSIL group,and SCC group were respectively.The expression level of miR-203 was lower than that of the NC group,and the difference was statistically significant(F=75.30,P<0.01).The expression of miR-203 between LSIL group and HSIL group and SCC group was significantly different(P<0.001).There was no significant difference in the expression of miR-203 between HSIL group and SCC group(P=0.174).And with the progression of cervical intraepithelial neoplasia lesions,miRNA-203 showed a significant downward trend.3.Spearman correlation analysis of miR-203 expression and folic acid content showed that miRNA-203 expression was significantly correlated with serum folic acid and positively correlated(r_s=0.836).The escalation of cervical intraepithelial neoplasia was linearly correlated with the decrease of serum folic acid and the low expression of miR-203(?~2=64.875,P<0.001).4.Ordered multivariate logistic regression analysis showed that the likelihood of cervical lesions graded at least one grade higher than the low-level low-expressing group,high-level high-expressing group,high-level low-expressing group,and low-level high-expressing group was 0.004(P<0.001),0.126(P=0.008),0.037(P<0.001).Conclusion:The abnormal expression of miRNA-203 and serum folate levels were closely related to the progression of cervical intraepithelial neoplasia.With the escalation of the degree of cervical intraepithelial neoplasia,the level of folic acid gradually decreased,and miRNA-203 was significantly down-regulated.Both showed significant changes.Positive correlation and linear correlation with cervical lesions,therefore,folic acid deficiency and low expression of miRNA-203 are high risk factors for cervical intraepithelial neoplasia.
