The hygroscopicity of Chinese herba preparation is one of the bottlenecks restricting in the development of traditional Chinese medicine(TCM).Solid preparation,in particular,most of them are made from extractum which is extracted from Chinese herba yinpian.As concrete is a more complicated composition system,containing some hygroscopicity of the component,such as polysaccharides,tannins,phlegm and a few small molecular alkaloids.These ingredients often leads to extract powder less liquidity,stronger viscosity,agglomerate,etc.This is also a important influencing factors in the process of preparation.In the process of storage,the hygroscopicity of composition also affects the stability and efficacy of solid preparation.Therefore,this subject mainly studies several moisture-proof technology that can obviously improve extract hygroscopicity of TCM to explore an ideal moistureproof method.According to the polysaccharide moisture absorption theory,this study choose the hygroscopic Astragalus polysaccharides(APS)as a model drug,using the technology of ion exchange,coating surface and reducing the specific surface area to research TCM extract moistureproof technology and establish a serise of moisture absorption characteristic parameters as indicators of moistureproof technology evaluation method and ultimately select the moistureproof method with better effect.By understanding moisture absorption principle and process,this study analyse and infer moistureproof mechanism of several moistureproof technologies.The main research contents and results are as follows.Through the aspects of physics,chemistry and Chinese medicine special properties,the author analyzes the reasons of TCM moisture absorption.Introducing all kinds of moistureproof technology for different reasons to Solve the problem of the moisture absorption of TCM from the raw material.With quadratic term mathematical model fitting material moisture absorption time curve,obtaining its model equation.After the model equation derivation,hygroscopic rate equation,initial velocity and accelerated velocity can be obtained,supplemented by equilibrium amount of moisture absorption and critical relative humidity as the moisture absorption characteristic parameters.This research adopts extract Astragalus polysaccharides as raw materials,Amberlite IRA-400 ion exchange resin as the carrier,resin drug loadings and the drug utilization as the evaluation index of process and speculates complex formation mechanism by differential scanning calorimetry(DSC)and infrared spectrometry(IF);Selecting the type of excipients and the proportion of drugs to excipients.Using mixture design method to optimize the proportion of excipients.Manufacturing polysaccharide-auxiliary materials mixed powder by auxiliary material modified method;Screening the kinds of water-soluble auxiliary material,through the mixture design method to optimize the ratio of excipients to manufacture the mixed powder of drug and excipients,Using it as raw material to research a granulating process,eventually,to make polysaccharide granules.Comparing the hygroscopicity of several moistureproof intermediates,the polysaccharide granules moistureproof effect is best.In addition,according to the formation mechanism of the resin compound I presume that it may have a potential to maintain the long-term stability of medicines.According to the principle and process of the moisture absorption,the research concludes damp-proof mechanism for several moistureproof technology.Auxiliary material modification method is through grinding and mixing auxiliary material with medicines,damaging the porosity and pore volume of the powder,hindering drug contact with outside air.Granulating technology mainly reduces the amount of porosity,which are exposed to the outside environment,by lowering the specific surface area;Through DSC and IF analysis,the ion exchange resin and APS form the resin composites by chemical bonding,not only physical adsorption.Therefore,the conclusion has been inferred that medicine is embedded and dispersed in ion exchange resin forming polymer by ion exchange technology,reducing the influence of external environment to medicines to enhance the stability of medicines.
