Chinese herbal preparation materials obtained by the pre-treatment process (Herbal Extracts) are complicated, mostly rendered illiquid, hygroscopic and other undesirable physical properties, such as extracts contain polysaccharides, tannins, alkaloids and other components of small molecules highly hygroscopic substance. The high hygroscopicity become a problem which long plagued Chinese medicine preparation and quality stability of solid formulations. The study select hygroscopic, illiquid Schisandra polysaccharide as a model drug, absorption parameters evaluation methods were established to study three kinds of modification technology in reducing the use of traditional Chinese medicine polysaccharides hygroscopic advantage and moisture-proof mechanism, the main research reads as follows:l.The extraction and purification technology research::The Schisandra polysaccharide extraction rate as the evaluation index, the use of composite design-response surface methodology preferred fructus schisandrae polysaccharide extraction process, and ultimately to establish the optimum extraction conditions of fructus schisandrae polysaccharide:refluxing temperature water bath100℃, solid-liquid ratio of1:41.48(g/mL), extraction time of5h, extracted two times. Deproteinized using Sevage law, after alcohol precipitation60℃drying the resulting light brown powder that was refined Schisandra polysaccharide.2.The establishment of moisture characteristic parameter evaluation methods:The moisture level of response time curves were obtained by analyzing the reaction equation level and hygroscopic coefficient; absorption time curve data for the regression analysis, the moisture binomial equation, after the initial moisture differential processing to calculate the speed, acceleration moisture, hygroscopic time this combination was able to better describe the behavior of traditional Chinese medicine extracts.3. Proof technology research:to study the surface of the powder coating, pelleting, the impact of these three modified microencapsulation technology Schisandra polysaccharide hygroscopic. The study found that different materials and process parameters will affect its moisture effects.4. Evaluation of the moistureproof effect of different modification techniques: moisture-time curve from the modified sample absorption coefficient of the first order reaction, moisture initial velocity, moisture acceleration, CRH, several aspects of mobility compared powder coated, granulating microencapsulated moisture modification of these three effects. Studies have shown that Schisandra polysaccharide by granulation optimal moisture effects after its CRH compared with unmodified Schisandra polysaccharide increased by10percentage points; samples for optimal mobility microencapsulated sample, its angle of repose of29.95°<30°.5. Moistureproof mechanism preliminary discussion:powder coated on the surface considered proof mechanism that was after the modification of fructus Schisandrae polysaccharide highly dispersed in carrier, the use of a carrier bag shielding effect of the polysaccharide hydrophilic groups in contact with water drops so as to achieve lower hygroscopicity effect; the moistureproof principle of granulation technique may be:the powder particles were prepared by, not only can improve the flowability, the specific surface area can also be reduced, and the time of extruding the extrusion granulation of the powder can destroy capillary tube, so as to achieve the purpose of reducing the moisture absorption; microencapsulation proof mechanism may be as follows:after microencapsulation, herbal extracts surface was covered in a layer of polymeric material to form a protective film, hindering the entry of moisture, which can the purpose to reduce hygroscopicity.
