| Objective:1.We intend to intervene the postmartum depression rat model by Chinese medicine shenguiren mixture.Observe the changes of behavior in rats.Detect the level of estrogen receptor(ERα、ERβ),brain neurotrophic factor(BDNF)and tyrosine kinase receptor(TrkB)mRNA protein expression in forntal lobe of PPD model rats.Explore the relationship among them.2.To investigate the prevalence of Postpartum Depression of parturient woman and analyse the related factors which influence Postpartum Depression.Methods:1.The mature female SD rats were ovariectomized bilaterally and received subcut-aneous injection of hormone(estrogen,progestin)for the model of postpartum depression.2.Using sucrose water consumption test,OFT,FST to conduct behavioral tests on animals after modeling.3.The rats were divided into 7 groups:(1)The model group(Mdl group),(2)Sertraline hydrochloride group(Ser group),(3)Estradiol benzoate group(EB group),(4)The shenguiren mixture of high dose group(S-H group),(5)The shenguiren mixture of medium dose group(S-M group),(6)The shenguiren mixture of low dose group(S-L group),(7)Control group(Ctl Group).4.In the second,fourth week after the intervention,conduct behavioral testing again.5.Measuring and analysis the protein expression of ERα、ERβ、BDNF、TrkB in frotal lobe of brin by RT-PCR.6.The sampling methods was non probability sampling.Selecting eligible parturient woman from Sichuan Provincial People’s Hospital and Sichuan Provincial Hospital of TCM who consented to participate were recruited into the study.The postpartum women enrolled were investigated by the Ediburgh Postnatal Depression Scala(EPDS).Conduct investigation and analysis the relevant factors which influence postpartum depression.Results:1.The establishment of postpartum depression model rats,behavioral index:compared with the blank control group,model group,rats in the Open Field Test scores,sucrose water consumption decreased(P<0.05),the swimming time shortened(P<0.05).2.Two weeks and a month,behavioral index:compared with the blank control group,model group,cane sugar consumption was lower(P<0.01),Open Field Test scores were significantly lower(P<0.01),the immobility time was significantly higher(P<0.01).Except Ser group,other groups,the sugar consumption was significantly higher than Mdl group(P<0.01);each Open Field Test scores was higher than Mdl group increased(P<0.01).Except the Ser group,other groups,swimming time were less than Mdl group(P<0.01)3.RT-PCR results:Compared with Ctl group,the expression levels of ERα,BDNF and TrkB in Mdl group were significantly lower(P<0.05),except for ER.Compared with Mdl group,Ser group,EB group,S-H group,SM group,S-L group,ERαmRNA expression increased,there was significant difference(P<0.05);the expression level of EB group,S-M group,BDNF TrkB mRNA increased,there were significant differences(P<0.05).4.In this study,a total of 289 questionnaires were collected,and a total of 282valid questionnaires were collected.The effective rate was about 97.58%.According to the diagnostic criteria,38 cases of maternal,postpartum depression prevalence rate was 18.45%5.The logistic regression analysis results show that the return coefficient of delivery mode B1=0.321,P1=0.490(P>0.05),the return coefficient of newborn sex B2=0.666,P2=0.155(P>0.05),there was no statistically significant difference.6.The Kendall,Spearman and Kruskal-Wallis H test results showed that postpartumdepressionassociatedwithmaternalageKendallcorrelation coefficient=0.017,bilateral P=0.741(P>0.05);Spearman correlation coefficient=0.020,bilateral P=0.742(P>0.05);Kruskal-Wallis H X2=0.717,P=0.699(approximate value P>0.05),there was no statistically significant difference.7.The incidence of postpartum depression did not decline with postpartum time,and even showed an upward trend,but there was no significant difference in the cumulative prevalence.Postpartum a week prevalence rate and postpartum a week after childbirth,postpartum a week for half a year to one year after childbirth and above stage prevalence,the Pearson%Χ~2 were 9.334 and 10.148,P were 0.004(P<0.05)and 0.003(P<0.05),there are differences statistical significance was not statistically significant compared to the rest of the time.Conclusion:1.PPD model rats can be successfully obtained by the method of hormone withdrawal after HSP.2.The shenguiren mixture can improve the depression behavior of PPD model rats.3.The shenguiren mixture may be able to further enhance the expression of BDNF and TrkB proteins by increasing the protein level of estrogen receptorαand thus play a role in anti depression.4.The prevalence of postpartum depression was 18.45%in this investigation.5.There was no correlation between the occurrence of postpartum depression and the mode of delivery,neonatal sex and maternal age.6.the incidence of postpartum depression may increase with the increase of postpartum time. |
