| Objective:By detecting the expression of Tyrosine kinase receptor B(TrkB)and brain derived neurotrophic factor(BDNF)in hypopharyngeal squamous cell carcinoma tissues(HSCC)and the effect of BDNF on the invasion ability of FaDu cell line,to analyze the relationship between the expression level of BDNF/TrkB and the clinicopathological characteristics of patients with HSCC and explore its relationship with the invasion and metastasis ability of hypopharyngeal carcinoma.Methods:The clinical data of 33 patients who were newly diag--nosed with HSCC admitted to the Affiliated Hospital of Jining Medical College from 2013 to 2016 with hypopharyngeal cancer were collected,and using immunohistochemical staining method to detect the expression levels of TrkB and BDNF in the cancerous tissues and paracancerous tissues of 33 patients with hypopharyngeal carcinoma and the normal pharyngeal mucosal epithelium of 12 non-cancerous patients.Using transwell invasion test to detect the effect of BDNF on the invasion ability of FaDu cell line.Results:1.Through the IHC we found that the expressions of BDNF and TrkB protein in hypopharyngeal carcinoma tissues are significantly higher than those of paracancerous tissues and normal pharyngeal mucosa tissues,they has statistical significance(P < 0.01);2.The expression of BDNF protein in the paracancerous tissues of hypopharyngeal carcinoma is significantly higher than that of normal pharyngeal mucosa tissue,it has statistically significant(P < 0.05);3.The expressions of BDNF and TrkB in hypopharyngeal carcinoma tissue samples are positively correlated,they have statistical significane(r=0.353;P=0.044,<0.05);4.The expressions of BDNF and TrkB protein in hypopharyngeal carcinoma tissue specimens are correlated with tumor diameter,degree of differentiation,lymph node metastasis,and clinical stage(P<0.05)and independent of gender,age and depth of infiltration(P>0.05).5.Through the Transwell invasion experiment we found that exogenous BDNF can significantly enhance the invasive ability of FaDu cells,The difference between the experimental group and the control group is statistically significant(P<0.05).Conclusion:1.The expression levels of BDNF and TrkB protein in hypopharyngeal carcinoma tissues are significantly higher than those in paracancerous tissues and normal pharyngeal mucosa tissues,and their expression levels are correlated with tumor diameter,degree of differentiation,lymph node metastasis,and clinical stage;2.BDNF can promote the invasion and metastasis ability of Fadu cell line. |
