Objective:Electroacupuncture"shengmen"and"sanyinjiao"to observing the effect of the behavior,adenylate activated protein kinase,acetyl-CoA,sodium and potassium triphosphatase and mitochondrial structure of hypothalamic paraventricular nucleus and plasma corticosterone insomnia of insomnia model rats,through electroacupuncture"shengmen"and"sanyinjiao",to explore the insomnia after fatigue of the energy metabolism of hypothalamic paraventricular nucleus.Method:66 healthy SD rats were randomly divided into blank group,model group and electroacupuncture group,each group of 22.The rats in the model group and the electroacupuncture group were modeled by restrictive braking for 15 days.After modeling,the electroacupuncture group electroacupuncture"shengmen"and"sanyinjiao"continuous treatment for 5 days.After modeling and treatment,respectively to experiment of 24 hours of spontaneous activity test,elevated cross labyrinth experiment,rat tail suspension experiment,negative xxhaustive swimming experiment,and at the end of treatment and behavioral testing to taken plasma and hypothalamic paraventricular nuclei,and to observing the expression of AMPK in the hypothalamic paraventricular nucleus,Ac-CoA and NA~+-K~+-ATP ase were detected in the mitral structure of the hypothalamic paraventricular nucleus and to measure the plasma CORT content.Results:(1)Using a restrictive braking method for 15 days,Compared with the blank group,the activity of spontaneous activity in the model group and the treatment group increased(P<0.05),nighttime activity decreased(P<0.05),the tail resting time increased(P<0.05),the elevated plus-maze test OE%and OT%were decreased(P<0.05),negative exhaustive swimming time is reduced(P<0.05).(2)Using a restrictive braking method for 15 days,Compared with the blank group,the expression of AMPK in the model group increased(P<0.05),the contents of Ac-CoA and NA~+-K~+-ATPase in PVN were decreased(P<0.05),plasma CORT content decreased(P<0.05),PVN mitochondrial structure appears partially changed.(3)Electroacupuncture"shengmen"and"sanyinjiao"treatment for 5 days,the treated rats 24h the amount of activities during the day were increased(P<0.05),the amount of night activities were decreased,and the time of tail suspension were shortened(P<0.05),elevated cross maze OE%and OT%were increased(P<0.05),exhaustive swimming time were Increased(P<0.05).There was a significant improvement in the behavioral indicatorsrecovery and close to normal level(P<0.05).(4)Electroacupuncture"shengmen"and"sanyinjiao"continuous treatment for 5days,the abnormal structure of the mitochondria of the treated rats was repaired,the contents of AMPK,Ac-CoA and NA~+-K~+-ATP ase,and plasma CORT were significantly improved(P<0.05),Close to normal group(P>0.05).Conclusion:(1)Insomnia rats have energy metabolism disorders,minifested as PVN mitochondrial morphological changes,the content of the AMPK expression increased,Ac-coA and Na~+-K~+-ATP ase decreased in PVN,and the plasma CORT content decreased.(2)Electroacupuncture"shengmen"and"sanyinjiao"acupoints can effectively improve the sleep state and fatigue symptoms of insomnia rats.(3)Acupuncturehasagoodadjustmenttorecoverytheenergy metabolism.Through intervention the effects of Ac-coA,NA~+-K~+-ATPase and AMPK expression in PVN,and plasma CORT.
