Objective:To investigate the psychological stress insomnia with Electric acupuncture rats Shenmen,after Sanyinjiao behavior and changes in blood and brain tissue Ach,5-HT,CRH,ACTH,CORT, and to explore the therapeutic effect of electro-acupuncture treatment for insomnia psychological stress in rats and mechanism of the HPA axis regulation.Method:Select30healthy SD rats were randomly divided into control group, model group and electro-acupuncture group,each group with10. Model group and electro-acupuncture group rats were restricted activity for15days’consecutive modeling. Conducted the day after modeling the elevated plus maze test,tail suspension test and24h spontaneous activity detection,model group and electro-acupuncture group were given acupuncture Shenmen.Sanyinjiao acupuncture treatment for4consecutive days. The control group.model group synchronous stationary treatment,elevated plus maze test performed after the treatment, and the tail suspension test24h locomotor activity test, and were tested post-test blood,brain tissue,respectively,of the Ach,5-HT,CRH, ACTH and CORT.Results:①Modeling three groups of rats in body weight,tail hanging inactivity, elevated plus maze OE%and OT%were no significant difference (P>0.05).②After three groups of rats modeling activity during the day,at night the amount of activity,elevated plus maze OE%and OT%has obvious difference (P<0.05),in which the model group and electro-acupuncture group,compared with the control group,the rats’ daytime activity had significantly increased,decreased in nocturnal activity.and elevated OE%and OT%reduced significantly (P<0.05). In the rat tail hanging,there still no significant difference in terms of time (P>0.05) after modeling the three groups of rats.③After electro-acupuncture three groups of rats have obvious difference in the daytime activity,nocturnal activity,elevated plus maze OE%and OT%(P<0.05).In spontaneous activities, model group compared with the control group significantly increased in daytime activity,significantly reduced in nocturnal activity,and electro-acupuncture group was significantly reduced in daytime activity compared with the model group while nocturnal activity increased significantly (P<0.05); In the elevated plus-maze test.the model group compared with the control group model group OE%and OT%significantly reduced.The electro-acupuncture group compared with the model group and the control group OE%and OT%increased significantly (P<0.05). And after electro-acupuncture treatment three groups of rats were no significant difference in the tail (P>0.05) static timing of the suspension.④After acupuncture treatment the plasma ACTH.CORT levels in three group rats have obvious differences (P<0.05). In plasma ACTH.the model group was significantly higher than the control group, while electro-acupuncture group was significantly lower than that in the model group,and significantly higher than the control group (P<0.05); In plasma CORT,the model group was significantly higher than the control group,and electro-acupuncture group compared with the model group was significantly lower (P<0.05),no significant difference compared with the control group(P>0.05).⑤After acupuncture treatment for three group rat in brain Ach,CRH levels with significant differences (P<0.05). In Ach, the model group was significantly higher than the control group,while electro-acupuncture group was significantly lower compared with the model group (P<0.05),no significant difference (P>0.05) compared with the control group; In CRH,the model group was significantly higher than the control group,while electro-acupuncture group was significantly lower compared with the model group (P<0.05),and no significant difference compared with the control group (P>0.05). And after electro-acupuncture group had no significant difference between the three5-HT levels in brain tissue of rats (P>0.05).Conclusion:The establishment of restrictive activity restraint stress insomnia rat model characterized by pathological features of sleep disorder and anxiety in behavior of significantly increase daytime activity,reduce nocturnal activity and decrease in OE%and OT%. Ach and CRH of Brain tissue in insomnia rat increased levels,and ACTH and CORT of plasma increased levels,HPAA hyperfuction. electro-acupuncture shenmen Sanyinjiao can reduce daytime activity and increase nocturnal activity,and elevated low OE%and OT%,in order to sleep well and relieve anxiety,which mechanism play therapeutic role through reducing the central Ach content to inhibiting excessive excitability of HPAA.
