This paper consists of two parts,the first part of Lycium ruthenicum Murr.quality standard were studied,the second part of the extraction and purification of pigment from Lycium rethenicum technology and antioxidant activity were studied.Lycium ruthenicum Murr.is a medicinal plant called Lycium chinense.It has abundant anthocyanin pigment.According to the records of "jingzhubencao" : its sweet,flat,with Yanggan Mingmu,kidney essence,Shengjinzhike effect etc.Modern pHarmacological research found that it has antioxidant,anti-aging,anti atherosclerosis and so on.At present,the standard system of the medicinal herbs has not been established.The purpose of this paper is to establish the quality standard of Lycium barbarum fruit in Qinghai Province,and to provide reference for the improvement of the quality standard of this herb.Including:(1)fruit base resource identification: for the first time in Qinghai Province,the main origin of the Qaidam Basin of Lycium ruthenicummurr Nuomuhong,Dulan,Delingha,and Golmud Xiangride several areas of Lycium ruthenicum were original plant identification;combined with literature research and field investigation,the Chinese growth environment the cultivation,processing and other areas have been revised.(2)the characters and microscopic identification of medicinal herbs: the characteristics and microstructure characteristics of Lycium barbarum were studied.(3)quantitative methods for the study of Lycium barbarum L.: a method for the determination of polysaccharides in betaine and pHenol sulfuric acid by HPLC-ELSD was established.The total anthocyanins,total polypHenols and proanthocyanidins in the medicinal materials were determined by spectropHotometry.(4)qualitative identification of Lycium ruthenicum Murr.: 13 batches of HPLC fingerprint was established,and Petunia-3-O-(p-coumaric acid)rutinoside-5-O-glucoside were studied TLC qualitative identification of traditional Chinese herbs.(5)the quality control of Lycium barbarum L.: the content of water,ash,extract and heavy metal of Lycium barbarum in different parts of Qinghai was determined and the limit was stipulated.(6)the establishment of the quality standard of Lycium barbarum fruit: formulated the quality standard for drafting Lycium barbarum and draft quality standard.Pigment ruthenicum(L.)is anthocyanin pigment.Modern pHarmacological studies showed that it has antioxidant and anti-aging activity.This paper uses a combination of BBD-response surface method of single factor test,the effect of percolation extraction of three main factors of ethanol concentration,percolation velocity,ratio of solvent were optimized.The selected X-5 macroporous resin had the best performance,the factors affecting the adsorption and desorption: sample concentration,sample size ratio,pH value,sample flow rate,water content,concentration of eluent,pH value,inspection agent flow rate elution,and the pigment value and yield of determination of.Results the optimum extraction process is the use of pH3.0,20 times the amount of 80% ethanol at a flow rate of 1mL/min for percolation extraction.Purification of pigment by X-5 macroporous resin,the ratio of diameter to height 1:15,pigment concentration 0.02g/mL,sample solution was pH3.0,the sample volume is 90 mL,a flow rate of 3 mL/min is the best adsorption condition;in 5BV 95% ethanol in pH4.0,flow rate of 3mL/min under the condition of best elution,purification of pigment yield was 7.33%,the color value is 21.7,good repeatability,and is suitable for industrialized production.The antioxidant activities of pigment extracts from Lycium barbarum were studied.The Lycium barbarum pigment has certain scavenging effects on DPPHfree radical,hydroxyl radical and superoxide anion radical.The scavenging ability of DPPH radical,hydroxyl radical and superoxide radical was better than that of VE,but lower than VC.
