Xiao'er Feiqingle Oral Liquid is an addition and subtraction formula on the basis of empirical formula of Chinese Academy of Traditional Chinese Medicine,which is confirmed effective of clearing heat and expectorant cough after years of clinical practice.The formula is composed by Honeysuckle,purslane,plantain grass,licorice.It mainly be used for treatment hyperactivity cough and dry mouth and sore throat,which are caused by the lungs fever.That is,western medicine,influenza,pneumonia,acute and chronic bronchitis,acute upper respiratory tract infection,acute pharyngitis,tonsillitis and so on.Common illness has cough,sputum,sputum yellow sticky,when there is fever,dry mouth sore throat,headache and so on.The topic change the original decoction compound to oral liquid on the basis of the basic theory of Chinese medicine and using modern formulation methods.In this paper,the preparation process,quality standard and stability of oral liquid were studied.According to the prescription of the herbs in the chemical composition and prescription efficacy,the optimum extraction parameters were obtained by using star point design-response surface method and single factor test with chlorogenic acid content and dry cream ratio as the index.The optimum extraction parameters are:Soak2h,make up the water absorption of 2.7 times,plus 10 times the amount of water reflux extraction 2 times,each 100 min.The five clarification methods of direct centrifugation,ethanol precipitation,chitosan flocculation,gelatin flocculation and activated carbon adsorption were compared by fingerprint and multi-index evaluation.The optimum process parameters of ethanol precipitation were as follows:the relative density of liquid was 1.2g / mL,the content of alcohol in the solution was50%,and the concentration of chlorogenic acid was calculated by the method of star point design and effect surface method,PH value of 5.0,frozen for 12 h.Through the study of preservatives and pH,determined to add no preservatives,finished pH value of 4.5 to 5.5.This article established the quality standard of Xiao'er Feiqingle Oral Liquid,finished product testing are in line with regulations.This paper established thequalitative test method of thin layer chromatography of honeysuckle,purslane,plantain grass and licorice.In addition,the finished fingerprints have been established.A method for the determination of chlorogenic acid in oral liquid was established.Specified oral liquid per 1mL not less than 7.5mg.Conducted a study on the stability of Xiao'er Feiqingle Oral Liquid,including freeze-thaw test,accelerated test and long-term test.Studies have shown that the preparation process is stable and feasible,quality control,can be used for industrial production guidance.
