Chinese Medicine Semen Cuscutae Treatment Of Knee Osteoarthritis Experimental And Clinical Research

Objective: To investigate the therapeutic effect of Chinese herbs Semen Cuscutae(SC)on early knee osteoarthritis;establish methods to choose a reasonable animal model,clear and definite the pathological stage;to evaluate the therapeutic effects of different SC extract on animal model of joints and screening of the potential role of monomer;looking for possible target joint repair induced by SC extract.Method:(1)20 healthy male SD rats were randomly divided into 5 groups(n = 4)in each group(n = 6).First,4,and 7 days after the experiment,the rats were anesthetized and injected into the right posterior articular cavity of to make the model of 0.1ml4% papain solution.The rats were sacrificed at second,3,4,6,and 8 weeks after modeling.The gross specimens were observed by naked eye,and the pathological changes of articular cartilage were observed under light microscope.(2)60 healthy male SD rats were randomly divided into 6 groups(n = 10)in each group(n = 6).In addition to the blank control group,the other groups were injected with the same volume of normal saline.Model fourth weeks according to the experimental plan for the corresponding drug intervention.During the period of observation,we recorded the changes of joint and measured the joint activity.At the end of 4 weeks after the intervention,the patients underwent X.After death,the gross specimens were observed by naked eye and scored,and the HE staining of articular cartilage was observed under light microscope to improve the Mankin s score.(3)By using the simple random method,according to the spss22.0 software to generate the random number table,60 cases of knee osteoarthritis patients receiving outpatient department of orthopedics were divided into experimental group(Chinese herbs SC free Decoction)and control group(oral glucosamine hydrochloride capsules),30 cases in each group.Two groups were treated for up to 8 weeks.Before and 4 weeks after treatment,after treatment WOMAC score and knee joint activity measurement.Finally,the data were analyzed statistically.Results:(1)in this experiment,the KOA model of SD rats was induced successfully by using the combination of 4% papain.(2)in the model,the animalsentered the stable KOA early stage in third week,and in the middle stage of KOA in the eighth week.(3)SC extract can effectively improve rat knee osteoarthritis cartilage degeneration,the effect was stronger than glucosamine,for the treatment of rat KOA specific effect.(3)SC polysaccharide plays a major therapeutic effect on rat KOA.Equal treatment dose SCPS is better than glucosamine,therapeutic effect of SC Flavonoids has not been confirmed(5)the clinical observation proved in improving the prognosis of patients with KOA,SC free decoction is stronger than glucosamine,faster onset,can think SC has certain advantages in the prevention and treatment of KOA.
