Objective:Obesity is a chronic metabolic disease with a very high incidence,and its formation is closely related to intestinal mucosal immunity,chronic inflammatory reactions,and intestinal flora abnormalities.This topic aims to study the effect of electro-acupuncture on the intestinal mucosal immunity in obese mice by replicating classic nutritional obesity animal models,and provide scientific basis of TCM to prevent and treat obesity.Method:Twenty-eight week-old SPF C57BL/6 mice were divided into males and females,and they were fed for one week.Randomly divided into normal control group 10,model group 110.The normal group was fed with standard diets,the model group was fed with high-fat diet of two months,and the body weight was more than 20%(or more than the normal group mean + 2 standard deviations)of the normal body weight of the same period.Obese mice with successful modeling were randomly divided into model group,acupuncture 7d group(A7),acupuncture 14 d group(A14),acupuncture21 d group(A21)and acupuncture 28 d group(A28),8 mice in each group.Weights were measured weekly and general signs and symptoms were recorded.Samples were taken on the 1st,2nd,3rd,and 4th week of treatment,and weighed.Blood was collected from the femoral artery.Serum was separated and blood glucose(GLU),total cholesterol(TC),high-density lipoprotein(HDL-C),and serum were measured.Density lipoprotein(LDL-C).Caesarean sections were taken from two to two sections of ileum midgut tissue,fixed in 10% neutral formaldehyde and stained with HE.The distribution density of Claudin-1 protein was analyzed by immunohistochemistry.Another ELISA was used to detect the changes of interleukin-2(IL-2),interleukin-10(IL-10),chemo kine(CC motif)ligand 28(CCL28),and regenerating islet-derived protein 3?(Reg3?).The expression of CCL28 mRNA and Reg3? mRNA was detected by RT-PCR.Result:Compare with normal mice,body weight,GLU,TC,HDL-C,and LDL-C serumconcentrations in obese mice were significantly higher(P<0.05).After electro-acupuncture treatment,the content of GLU in group A7 decreased significantly,and total cholesterol and high-density lipoprotein decreased significantly in each group(P<0.05).The results of HE staining showed that the intestinal mucosa structures in the normal mice was intact,and the intestinal mucosal epithelial cells of the obese mice were swollen and degenerated,shed,part from the intestinal gland necrosis,central chymous disappeared,submucosal edema,and inflammatory cells infiltrated.The degree of intestinal mucosal lesions improved after electro-acupuncture treatment.The distribution density of Claudin-1 protein in the jejunum of obese mice was decreased(0.2086±0.0052).After electro-acupuncture treatment,the distribution density of Claudin-1 protein was significantly improved on the A14(0.2347±0.0180)group(P<0.05).Obese mice had serum IL-2(108.7±21.72 VS 238.66±27.78),IL-10(114.45±15.93 VS 278.66±39.88),and Reg3 ?(352.28±47.91 VS 734.68±49.81)compared with normal mice.The content was significantly decreased(P<0.05);the protein content of Ccl28(385.26±50.73 VS177.87±36.17)was significantly higher(P<0.05).After acupuncture treatment,the levels of IL-2,IL-10 and Reg3? increased in each group(P<0.05),and the protein content of Ccl28 decreased(P<0.05).The expression of Ccl28 gene in obese mice was higher than that in normal group(1.91±0.45 VS 1.01±0.16)(P<0.05),but the expression level of Reg3? gene was significantly decreased(0.35±0.04 VS 1.01±0.16)(P<0.05).However,after electro-acupuncture treatment,the expression of Ccl28 gene gradually decreased(P<0.05),and the expression of Reg3 ? gene increased slowly(P<0.05).Conclusion:Electro-acupuncture has a significant effect on the treatment of obesity.Adjusting intestinal mucosal immune function of obese mice is one of the most important mechanisms.
