Objective:In this experiment,the effects of different ratios of Ferulic acid and astragaloside A on the tension of thoracic aorta of rats were studied.The mechanism of relaxation of thoracic aortic rings by different agonists and antagonists?Ferulic acid,FA?and astragaloside ??AS ??.Methods:The changes of tension of isometric thoracic aortic rings were collected by tissue perfusion system and powerlab data processing system in vitro.Ferulic acid was observed and compared with different concentrations of ferulic acid.The effects of astragaloside ?,ferulic acid and astragaloside ? on the tension of thoracic aorta in rats were studied.To observe the effect of ferulic acid and astragaloside on the tension of vascular ring in rats,and compare the difference between the two groups.The effects of ferulic acid and astragaloside ? on the relaxation of thoracic aortic rings induced by different agonists were observed.To observe the effects of different potassium channel blockers Tea,4-Ap,Gly,Bacl2,nos inhibitor L-nameand,cox inhibitor Indo on vasodilation of ferulic acid and astragaloside.Results:1.The effects of intravenously on the tension of thoracic aorta in rats under resting conditionThe experimental results showed that the cumulative addition of1×10-4-3×10-1.5g/L iv had no significant effect on the thoracic aorta of rats in basal state,but there was no significant difference compared with the solvent control group?p>0.05?.2.The effects of titration on thoracic aorta tension in rats with hyperkalemia The results showed that ferulic acid was added on the basis of KCL precontraction.Astragaloside ?(1×10-4-3×10-1.5g/L)could dilate thoracic aorta FA in a concentration-dependent manner.The maximal relaxation force of Astragaloside? on the rings was 73.24%±7.69,58.36%±4.41%,which was significantly different from that of the solvent control group?23.16%±2.35%?P<0.001).3.Effects of different ratios of FA and AS ? on the tension of thoracic aorta in high potassium stateThe results showed that on the basis of kcl preconstriction,the maximal diastolic rate of thoracic aortic rings in different ratios was 96.63%±4.68%?67.59%±4.85%?46.27%±5.14%?90.35%±4.74%?78.51%±5.37%respectively.There was significant difference between the two groups?73.24%±7.69%?and 1:1 group?1:10?.The maximal relaxation rate was compatibility of Astragalus membranaceus was 1:5.We compared with different compatibility with 1:5 Emax of 67.59%±4.85%,and found that there was a significant difference compared with 1:1 group,5:1 group,1:10 group.The diastolic rate was 96.63%±4.68%?90.35%±4.74%?78.51%±5.37%respectively.The results showed that these three ratios were obviously superior to the classical formula,but the diastolic rate of 1:1 was the most obvious,so we further studied the mechanism represented by 1:1.4.Effect of FA+AS ?1:1 on tension of thoracic aortic rings in ratsThe results showed that the cumulative ratio 2.654±0.28 and67.43%±5.07%,EC500 2.198±0.33?P<0.01?respectively.The results suggest that the 1:1 ratio of ferulic acid and astragaloside A in vasodilator depends on the receptor-operable Ca2+channel(Ca2+channel)and voltage-dependent Ca2+channel?VDC?.6.The effect of FA+AS ?1:1 on the tension of thoracic aortic rings in rats with K+channel blocker Tea,4-Ap,Gly,Bacl2The results showed that tea,a K+channel blocker,which was dependent on the contraction of thoracic aortic rings by kcl,could significantly inhibit the concentration-dependent vasodilation of 1:1(1×10-4-3×10-1.5g/L).The emax of thoracic aortic rings in rats was 31.7%±3.91,which decreased 35.73%±2.08compared with the control group,and the voltage dependent K+channel blocker4-Ap could significantly attenuate the relaxation effect of ferulic acid and astragaloside1:1(1×10-4-3×10-1.5g/L).The Emax of thoracic aorta in rats was41.39%±7.41,which was decreased by 25.64%±1.44%compared with the control group.Gly,a K+channel blocker?P<0.001?,could inhibit the contraction of thoracic aortic rings induced by hyperkalemia.Its Emax was 54.92%±6.95,which was 12.51%±1.25 lower than that of the control group,and EC500 was 1.648±0.05.The inward rectifier K+channel blocker Bacl2 could inhibit the diastolic Emax of ferulic acid and astragaloside1:1(1×10-4-3×10-1.5g/L)by 50.25%±10.52%,and decreased by 15.19%±4.55%?p<0.01?,and EC500 was 1.713±0.28compared with the control group.The above results suggest that the vasodilative effect of Iv1:1 may be related to the opening of KIRKATPTP channel,which was decreased by 25.64%±1.44%compared with the control groupglya K+channel,1.39%±7.41%,which was decreased by 25.64%±1.44%compared with the control groupglya K+channel.7.Effect of FA+AS ?1:1 on the tension of thoracic aortic rings in rats under the condition of L-name,IndoThe results showed that on the basis of Phe?1 mol/l?precontracted thoracic aorta ring,nitric oxide synthase NOS inhibitor L-NAME and COX inhibitor Indo could inhibit the diastolic effect of astragaloside and ferulic acid ratio 1:1 on rat thoracic aorta ring,the Emax was 51.86±12.07%,55.11%±6.95%respectively,and decreased compared with the control group,respectively.The results were29.65%±7.79%,30.4%±2.43%?P<0.001?,and the results of EC500 were1.708±0.31,1.693±0.29respectively.The results showed that no and PGI2 were involved in the vasodilatation of FA+AS ?1:1.Conclusion:1.The main components of Danggui Buxue decoction,ferulic acid,astragaloside A and their different compatibility have different degrees of diastolic effect on vascular rings.When the compatibility ratio changed,the diastolic rate of vascular rings was also changing.Compared with different ratios and monomers,the 1:1 diastolic rate of ferulic acid and astragaloside I was the highest,that is,when the proportion of ferulic acid or astragaloside in the mixture gradually increased,On the contrary,the relaxation rate of vascular rings showed a decreasing trend,indicating that different compatibility had different diastolic characteristics.In the study of the comparison of monomer and different compatibility,it was found that the relaxation effect of different proportion of compatibility was better than that of monomer.2.The 1:1 ratio of ferulic acid and astragaloside A had a relaxing effe ct on the thoracic aortic rings.The endothelium-dependent and non-endothelium-dependent pathway was associated with the inhibitory effect of L-name on the relaxation of thoracic aortic rings at 1:1 ratio of ferulic acid and astragaloside.Indo,an inhibitor of cyclooxygenase,also inhibited the v asodilative effect of ferulic acid and astragaloside ? at 1:1,indicating that PGI2 was also involved in the relaxation of aortic rings.The results show ed that the vasodilating effect of ferulic acid and astragaloside ? at 1:1 w as related to the effect of no on the relaxation of thoracic aorta in rats aft er preincubation with teaanine 4-Ap?Tea?Gly?Bacl2,a channel blocker.T he vasodilating effect of ferulic acid and astragaloside A at 1:1 may be rel ated to the kvkcai kirkirkatp channel,and its cellular mechanism may be re lated to the inhibition of internal Ca2+release by blocking the inward flow of Ca2+by blocking VDC and ROC.
