Objective To investigate the effect of Astragaloside ? and Ferulic acid on the human mesangial cells,and to investigate the intervention effect of Astragaloside ? and Ferulic acid to the expression of Smad,Smurf2 in human mesangial cells.Methods The cultured cells were divided into normal glucose(NG group,5.6mmol/L),high glucose(HG group,30mmol/L),high glucose plus Astragaloside ? at 1,10,100 umol/L definition in AS1 group,AS 10 group and AS 100 group,high glucose plus Ferulic acid at 1,10,100umol/L definition in FA1 group,FA 10 group and FA 100 group,and high glucose plus Astragaloside ? 1 umol/L and Ferulic acid 1 umol/L definition in A1+F1 group.HMCs proliferation ability in 24,48,72h was detected by MTT colorimetry.Collect HMCs of all groups at 48h,the expression of Smad2,Smad7 and Smurf2 mRNA were observed with using Real-time PCR.Collecting HMCs of all groups at 48h,TGF-?1,Smad2,Smad7,Smurf2 levels were measured with ELISA.Results 1.The cell proliferation of HG group was increased compared with NG Group(p<0.01).All medication Groups showed a significant decrease of HMCs proliferation in concentration-dependent manners compared with HG group(p<0.05,p<0.01),but still higher than NG group.2.mRNA expression:?Compared with NG Group,the expression of Smad2,Smurf2 mRNA significant increased in HG group(p<0.01),and Smad7 mRNA decresed(p<0.01).?Compared with HG Group,Smad2 mRNA lever showed a significant decreased(p<0.01).No change could be found between HG and A1+F1 groups.?The expression of Smurf2 mRNA decreased except FA1 group(p<0.01).?Reduced Smad7 mRNA in HMC of HG were elevated slightly after 100 umol/L Astragaloside ? administration(P<0.01).There were no change in other medcine groups compared with the HG group.3.TGF-?1 expression:the expression of TGF-?1 was increased significant in HG group compared with NG Group(p<0.01).After treatting HMCs with medication,TGF-?1 lever was decreased(P<0.05,P<0.01).But still higher than NG group except FA100 group.4.The level of Smad2,Smad7,Smurf2:? Smad2,Smurf2 lever in HG group increased(P<0.01),the expression of Smad7 decrased(P<0.01).?As the concentration increases,the expression of Smad2,Smurf2 protein in all medication groups reduced compared with HG group(P<0.01)except FA1 group.?The lever of Smad7 increased in concentration-dependent manners after tratmenting HMCs(P<0.05,P<0.01)except for AS1,FA1 group,but still higher than NG group(P<0.01).?The Smad7 protein were negative to Smurf2 protein(p<0.01).Conclusion 1.Astragaloside ? and Ferulic acid could inhibit HMCs proliferation under high-glucose.They reduced the lever of TGF-?1 and Smad2,increased Smad7.Astragaloside ?and Ferulic acid could increase the Smad7 protein by decreasing the lever of Smurf2,which could reduce the degradation of Smad7 via ubiquitination.2.The combination of Astragaloside ? and Ferulic acid in low concentrations shown significant synergy effect.
