Objective:? Investigate the cerebral palsy twins and single birth of combination disease and syndrome to analyze the high risk factors and syndromes distribution characteristics in children with cerebral palsy.? Use the epigenetics method of DNA methylation to discuss in the study of syndromes in children with cerebral palsy.Methods:? The theoretical study of cerebral palsy:the research status of cerebral palsy in China during the last 40 years was analyzed by CNKI database,and the advantages and significance of using double offspring materials to study cerebral palsy were discussed.? Study on the combination of disease and syndrome of cerebral palsy:The clinical questionnaire of etiology of cerebral palsy in children,the questionnaire of TCM syndromes of children with cerebral palsy and the expert diagnosis were used to investigate the general situation and syndromes of the selected twins of cerebral palsy and single fetus of cerebral palsy,and to analyze the etiology of cerebral palsy.Risk factors and syndromes distribution.? Methylation study of cerebral palsy:The HumanMethylation450 BeadChip(450k methylation chip)technique was used to scan the whole blood DNA methylation spectrum of 2 pairs of "normal cerebral palsy" twins,7 cases of cerebral palsy single birth and 1 healthy children,and screening the twins,the 1 twins normal single fetus,the 2 twins normal single fetus,the 1 twins cerebral palsy single fetus,2 to 2.The differential methylation sites of twins,cerebral palsy,single fetuses and normal single fetuses were analyzed.The difference methylation sites were analyzed by GO and KEGG database.Results:? The result of the disease syndrome:the risk factors of children with cerebral palsy were prenatal maternal factor,hyperarousal,Anorexia,Low birth weight,premature birth,anoxia,etc.Among the 120 children with cerebral palsy,the risk factors of 108 children with cerebral palsy were two or more.The results of the syndrome were mainly spleen and kidney deficiency,and the severity of the disease was different in the twins.? Results of methylation study in children with cerebral palsy:According to the GO and KEGG database analysis,the occurrence of cerebral palsy involves a large number of abnormal methylation sites.The difference methylation sites are more annotated by the KEGG database than the immune related pathways.The results of the GO database function annotation are more than the immune and neurodevelopmental classes in the BP layer,and are more related to the transport mechanism in the CC layer than in the CC layer,and in M The F layer is related to the enzyme activity.Conclusion:?Spleen and kidney deficiency play a leading role in the syndrome of cerebral palsy.?The pathogenic factors of cerebral palsy are complex,the risk factors are more than two kinds,and the severity is different.?The greater the difference in MZ between children with cerebral palsy,especially between identical twins,the greater the number of differential methylation sites.? The occurrence of cerebral palsy involves a large number of abnormal expression of methylation sites,especially the immune related GO and KEGG pathways.
