Objective:To discuss the effect of the method of Tonifying Kidney and Removing Phlegm?TKRP?through APN/AMPK pathway on reproductive hormone disorder,follicular dysplasia and insulin resistance?IR?in obese rats with polycystic ovary syndrome?PCOS?,and to preliminarily discuss the molecular mechanism of the method in the treatment of obese PCOS.Method:To select SPF female SD rats aged 7 to 8 weeks,besides blank control group,the other rats randomly divided into six groups:model control group,high dose group of traditional Chinese medicine?TCM?,middle dose group of TCM,low dose group of TCM,and dain-35 group after 28 days of intragastric administration of Letrozole and hyperlipidemia suspension.The high,meddle and low dose groups of TCM were given equal volume of traditional Chinese medicine,the dain-35 group was given equal volume of dain-35 solution,the blank control group and the model control group were given equal volume distilled water,respectively.To calculate body weight growth rate,and detecte the content of serum reproductive hormone E2?T?LH?FSH by radioimmunoassay and calculate the LH/FSH.The content of fasting blood-glucose?FBG?was detected by enzyme assay,the content of serum adiponectin content and fasting insulin?FIN?were detected by Elisa,and calculate the HOMA-IR.The expression of AdipoR1?INR?GLUT4 in granulosa cells?GC?were detected by immunohistochemistry,and the expression of AMPK mRNA?CYP19a mRNA in GC was detected by Realtime PCR.Result:1.Compared with the model control group,the body weight growth rate,the content of serum T?LH?FBG?FIN?LH/FSH?HOMA-IR of high dose group and middle dose group were decreased significantly?P<0.05?.2.Compared with the model control group,the content of serum E2?FSH?APN?the expression of AdipoR1?INR?GLUT4 and the relative expression of AMPK mRNA?CYP19a mRNA in GC increased significantly?P<0.05?.3.APN was positively correlated with AdipoR1 and AMPK mRNA?P<0.05?.APN and AMPK mRNA were positively correlated with CYP19a mRNA?E2?GLUT4?P<0.05?,and were negatively correlated with T?LH/FSH?HOMA-IR?P<0.05?.HOMA-IR was negatively correlated with E2?CYP19a mRNA?P<0.05?,and was positively correlated with body weight growth rate and serum T?P<0.05?.GLUT4 was positively correlated with E2?CYP19a mRNA?P<0.05?,and was negatively correlated with body weight growth rate and serum T?P<0.05?.Conclusion:1.The inhibition of APN/AMPK pathway in obese PCOS may be the mechanism of inducing IR and hyperandrogen.2.The method of TKRP can reduce the body weight growth rate and improve the reproductive endocrine disorder and IR by activating APN/AMPK pathway.3.The method of TKRP can effectively treat obese PCOS. |
