Objective: To choose the extract of Qi Deng Mingmu capsule,the traditional Chinese medicine of benefiting qi for activating blood circulation and nourishing Yin,and exploring its effect on the retinal neovascularization induced by oxygen in mice and the related mechanism,so as to provide experimental basis and new research ideas for the clinical treatment of retinal neovascular diseases with traditional Chinese medicine.Methods: 53 newly born C57/6J mice(male and female)were treated with hyperoxia induced retinal neovascularization model.The mice were divided into model group and normal control group.The model mice were kept in the oxygen concentration(75± 2%)environment for 5 days and returned to the normal air for 5 days,and the normal exposure group was kept in the normal air.In the two groups,at 17 days of age,7 mice in each group were taken pathological sections of retina,retinal angiography and retina paving.The changes of the retinal neovascularization in the two groups were observed and the morphological changes of retinal vessels were observed.After the success of the model,31 model mice were randomly divided into the model group,the low dose group,the middle dose group and the high dose group,6 in each group,7 in the apatinii group.The apatinib group was used to inhibit the70mg/kg of the neovascularization drug apatinib in the mice for 17 days,and the Qi Deng Mingmu capsule was used as a low,medium and high dose.On the 17 day of the mice,the rats in the group were gavaged with Qi Deng Mingmu capsule,and the dosage was low(200mg/kg),medium(400mg/kg)and high dose group(800mg/kg).In addition to the normal control group and the model group,the mice in the rest of the mice were continuously fed for 1 weeks,and the mice were killed after 1 weeks.The expression of nerve cilium 1(NRP-1)and endothelial growth factor receptor 2(VEGFR-2)in the retina tissues of each group was detected by immunohistochemistry;Western blot was used to analyze the retina retina tissue.Expression of middle galactoglutinin 1(GAL-1)protein.The results were analyzed statistically.Results: The retinal vasculature and the retina spread showed that the retinal vascular circuitous expansion,fluorescein leakage and the non perfusion area of the retina were found in the model group.The retinal vessels in the normal irradiated mice were evenly distributed,no fluorescein leakage and no perfusion area were found.In the pathological section of retina,the number of endothelial cells in the inner limiting membrane of the model group was higher than that in the model group was15.6000±5.9414,the normal control group was 5.6000 ±2.3022,and the two groups had significant difference P<0.01.Compared with the normal control group,the content of NRP-1 and VEGFR-2 protein in the eyeball tissues of the model mice increased significantly,and had significant statistical significance(P<0.01).Compared with the model group,the content of NRP-1 protein in the high dose group of apatinib group,the Qi Deng Mingmu capsule xtract and the middle dose group of the mice was significantly lower,with significant statistical significance(P<0.01).The content of NRP-1 protein in the retina of the control group was decreased,which was statistically significant(P<0.05).Compared with the model group,the content of VEGFR-2 protein in the retina tissue of the high dose group of apatinib group and Qi Deng Mingmu capsule extract decreased and had statistical significance(P<0.05).The Western blot GAL-1 results showed that the expression of GAL-1 protein in the retina tissue of the model mice increased significantly(P < 0.01)compared with the blank group(P < 0.01).The expression of GAL-1 protein in the retina tissue of the mice was reduced significantly(P < 0.01),and the expression of GAL-1 protein in the retina tissue of high dose group of Qi Deng Mingmu capsule was lower(P < 0.05).Conclusion: The use of Qi Deng Mingmu capsule extract,which has the efficacy of benefiting qi for activating blood circulation and nourishing Yin,can regulate the expression of hyperoxia induced retinal neovascular growth factor and improve the structure and function of retina.It is presumed that it has a certain protective effect on the visual function of patients with retinal neovascularization disease.
