Objective:To investigate the relationship between the gene polymorphism of CETP,APOA1 and metabolic syndrome(MS)in Uyghur,and further analyse the gene-gene interaction,which can provide theoretical basis for the prevention and treatment of MS in Uyghur.Methods:1.A number of 282 patients with MS and 298 controls were randomly selected from the reliminary database after matching the gender.We detected the CETP and APOA1 polymorphisms by usying the SNaPshot method.2.The basic statistical analysis was carried out by SPSS17.0,and the relationship between the polymorphisms of CETP and APOA1 genes and MS was analyzed by using logistic regression,t test, and Chi-square test.3.SHEsis software was used for Hardy-Weinberg test,linkage disequilibrium and the construction of haplotype.4.Multifactor dimensionality reduction(MDR)was used to analyze the influence of gene-gene interaction on MS among the Uyghur.Results:1.The rs4783961 did not conform to the Hardy-Weinberg balance(P<0.05).2.Significant different was found on the distribution of rs3764261,rs1800775,rs708272,rs289714,rs12149545 and rs711752 genotype between two groups(all P<0.05).The allele frequencies of rs3764261-T,rs708272-T,rs12149545-A and rs711752-A were lower in case group(all P<0.05),while the frequencies allele of rs1800775-C and rs289714-C were higher than those in case group(all P<0.05).3.Rs3764261-GT/TT,rs708272-CT/TT,rs12149545-GA/AA and rs711752-AA genotypes in CETP might reduce the risk of MS in the Uyghur population(P<0.05).4.Significant different was found on the distribution of rs5072,rs7116797 and rs2070665 genotype between two groups(all P<0.05).The allele frequency of rs670-A,rs5072-T and rs2070665-T was higher in the case group(all P<0.05).5.Rs5072-CT,rs7116797-AG and rs2070665-CT genotype in APOA1 might increase the risk of MS in the Uyghur population(P<0.05).6.Complete linkage disequilibrium was identified for two pairs of single nucleotide polymorphisms (rs3764261 and rs12149545(D'=1.000,r~2=0.929),rs711752 and rs708272(D'=1.000,r~2=0.997).The G-A-C-G-G(P=0.017,OR=0.635,95%CI=0.437-0.922)and T-A-T-A-A(P<0.001,OR=0.510,95%CI=0.381-0.684)haplotypes might reduce the risk of MS.7.Complete linkage disequilibrium was identified between rs5072 and rs2070665(D'=1.000,r~2=0.996).The T-A-T(P=0.017,OR=0.727,95%CI=0.559-0.945)haplotype might reduce the risk of MS.8.Interaction analysis of CETP gene showed that all the interaction models without statistical significance (all P>0.05).9.Interaction analysis of APOA1 gene showed that the best model was four order interaction model (rs670-rs5072-rs7116797-rs12718464)(P=0.017,CV Consistency=10/10).10.Interaction analysis of CETP and APOA1 gene showed that the best model was six order interaction model(rs3764261-rs1800775-rs5882-rs708272-rs289714-rs7116797)(P=0.017,CV Consistency=10/10).Conclusions:1.Rs3764261,rs1800775,rs708272,rs289714,rs12149545 and rs711752 were associated with the risk of S.The rs3764261-GT/TT,rs708272-CT/TT,rs12149545-GA/AA and rs711752-AA genotypes might e protective factors of MS.Rs1800775-CC and rs289714-CC genotypes were risk factors of MS.2.The rs670,rs5072,rs7116797 and rs2070665 polymorphisms were associated with the risk of MS.The s5072-CT,rs7116797-AG and rs2070665-CT genotype might be the risk factors of MS.3.Complete LD was found between two pairs of SNPs(rs711752 and rs708272,rs3764261 and s12149545).The G-A-C-G-G and T-A-T-A-A haplotypes might be protective factors for MS among the yghur.4.Complete LD was found between rs5072 and rs2070665.The T-A-T haplotype might be a protective actor for MS among the Uyghur.5.No interaction existed among CETP gene polymorphisms.6.Interaction was found among the APOA1 gene polymorphisms.The best interaction model was four rder interaction model(rs670-rs5072-rs7116797-rs12718464).7.Interaction was found between CETP gene and APOA1 gene polymorphism.The best interaction model as six order interaction model(rs3764261-rs1800775-rs5882-rs708272-rs289714-rs7116797).
