Objective:To direct and compare the serum as well as genetic level of 14-3-3 eta protein in patients at different rheumatoid arthritis(RA)stages with control group subjects,then to analyze the relationship between them and RA related laboratory and clinic indexes,separately,so as to investigate the significance of 14-3-3 eta protein to RA diagnosis and disease activity.Subjects and Methods:A total of 59 patients with RA including 19 preliminary diagnosed cases,20non-preliminary diagnosed cases and 20 stable cases,as well as 20 healthy control subjects were enrolled.Compare the serum and genetic level of 14-3-3 eta protein between these four groups by euzymelinked immunosorbent assay and Real-time fluorescence quantitative PCR,respectively,and then analyze the relationship in RA patients between 14-3-3 eta protein and C-reactive protein(CRP),erythtocyte sedimentation rate(ESR),rheumatoid factor(RF),anti-cyclic citrulline protein antibodies(anti-CCP antibody),age,disease course,time of therapy,tender joint count(TJC),swollen joint count(SJC),visual analogue scale(VAS)and disease activity score(DAS-28).Statistical analysis was performed with Wilcoxon rank sum test,analysis of variance,Chi-square test,Kruskal-Wallis,Pearson and Spearman correlation test.Results:1.The serum level of 14-3-3 eta protein in RA groups(1.68,0.63–2.89 ng/ml)is significantly higher than the control group(0.63,0.63–1.20 ng/ml,P<0.00).Both non-preliminary diagnosed group(1.65,0.63–3.72ng/ml)and stable group(0.63,0.63–1.94ng/ml)have lower serum 14-3-3 eta protein level than that of preliminary diagnosed group(2.58,1.65–3.66ng/ml),furthermore,the stable group has a lower serum 14-3-3 eta protein level than non-preliminary diagnosed group(P<0.05).2.The genetic level of 14-3-3 eta protein in RA groups(1.95+1.60)is significantly higher than the control group(0.92+0.19,P<0.01),and there is no significant difference between the preliminary diagnosed group(1.69+0.00),non-preliminary diagnosed group(2.32+0.98)and stable group(1.83+1.54,P=0.07).3.The relationships between serum level of 14-3-3 eta protein in 59 RA patients with their CRP,ESR,RF are all positive related(r=0.38,P=0.00;r=0.32,P=0.01;r=0.03,P=0.84,respectively),while the relationship with anti-CCP antibody(r=-0.02,P=0.90)is negative,but only the relationships of CRP and ESR are significantly different(P<0.05).4.The relationships between genetic level of 14-3-3 eta protein in 59 RA patients with their ESR,RF,anti-CCP antibody are all positive related(r=0.11,P=0.39;r=0.22,P=0.11;r=0.09,P=0.50,respectively),while the relationship with CRP(r=-0.01,P=0.92)is negative,but none of the relationships is significantly different(P>0.05).5.The relationships between serum level of 14-3-3 eta protein in 59 RA patients with their clinical indexes including age,disease course,TJC,SJC,DAS-28(CRP)and DAS-28(ESR)are positive related(r=0.07,P=0.59;r=0.38,P=0.00;r=0.43,P=0.00;r=0.41,P=0.00;r=0.50,P=0.00;r=0.43,P=0.00,respectively),while the relationship with therapy time is negative(r=-0.39,P=0.00),and all the relationships with disease course,therapy time,TJC,SJC,DAS-28(CRP)and DAS-28(ESR)are significantly different(P<0.05).6.The relationships between genetic level of 14-3-3 eta protein in 59 RA patients with their clinical indexes including age,disease course,therapy time TJC,SJC,DAS-28(CRP)and DAS-28(ESR)are all negative related(r=-0.14,P=0.29;r=-0.01,P=0.92;r=-0.13,P=0.31;r=-0.13,P=0.31;r=-0.14,P=0.29;r=-0.02,P=0.89;r=-0.08,P=0.54,respectively),but none of which is significantly different(P>0.05)..Conclusion:1.The onset of RA is related with its serum and genetic level of 14-3-3 eta protein.2.The combined usage of 14-3-3 eta protein,RF and anti-CCP antibody could help to diagnose RA more effectively.3.The serum level of 14-3-3 eta protein could be used as a new laboratory index to evaluate RA disease activity and could help to provide a treatment plan.
