Objective:To investigate the expression of Atox1 and Lox,and the effect of Valsartan on it after vascular balloon injury in rats.Exploring the role of Atox1 and Lox in the process of endometrial hyperplasia and the role of valsartan in inhabiting the restenosis after vascular injury.Methods:Thirty-six Wistar rats,weighing 300-350 g,were randomly divided into Control group?n=12?,Operation group?n=12?,and Drug group?n=12?.The model of restenosis after balloon injury of aorta was used in rats.The Control group was not given balloon dilatation and was given normal saline intragastric perfusion since one day before operation;the Operation group was given balloon dilatationand was given normal saline intragastric perfusion since one day before operation;the Drug group was given balloon dilatation,and the valsartan 20 mg kg-1d-1was given intragastric perfusion from the first day before the operation.Three groups of rats respectively on postoperative 14th day,28th day respectively take its aorta,paraffin wax slices after HE staining,morphology changes of tunicaintima were observed under light vascular,and computer image analysis system to detect the aortic intima,the film thickness.By real-time fluorescent quantitative PCR detection Atox1 in the vascular tissue and Lox mRNA expression,immunohistochemical detection Atox1 protein expression,and the use of image analysis software to the accumulative optical density value indicates that the relative content of the protein,using enzyme-linked immunoassay detection Lox level.SPSS17.0 was used for data processing,and the results of the measurement data were expressed by x±s.The variance analysis was used to compare multiple groups,and LSD-t was used for comparison between groups,and P<0.05 was considered statistically significant.Results:?1?After balloon injury,the thickness of intima and media of aorta increased significantly in the Operation group,and there was significant difference between the two groups?P<0.05?.The expression levels of Atox1,Lox m RNA and protein in aortic vessels of the Operation group were significantly higher than those in the Control group?P<0.05?.?2?Compared with the Operation group,the proliferation of intima and media of aorta decreased significantly?P<0.05?,and the expression of Atox1,Lox mRNA and protein decreased in the Drug group after valsartan administration.Compared with the Operation group,the difference between the two groups was statistically significant?P<0.05?.Conclusion:?1?In rats with aortic balloon injury,intimal hyperplasia was obvious,and the expression of Atox1 and Lox increased,suggesting that Atox1 and Lox may participate in the process of postoperative stenosis of vascular balloon injury.?2?After the use of valsartan endometrial hyperplasia,the expression of Atox1 and Lox decreased,indicating that valsartan could inhibit the restenosis after the injury of vascular balloon,and the mechanism may be related to the inhibition of Atox1 and Lox.
