Objective:To explore the relationship between PPAP2B gene polymorphism and cerebrovascular stenosis.Furthermore,the interaction between PPAP 2B gene and environmental risk factors was explored to provide clues for the genetic study of cerebrovascular stenosis.Methods:170 patients with cerebral vascular stenosis and 130 healthy volunteers(control group)who were treated in department of neurology,affiliated hospital of Qingdao university from March 2016 to October 2017 were selected.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)technology was used to detect the genotypes of two SNP sites(rs 17114036 and rs 6588635)of PPAP 2B gene.Using hardy-Weinberg law to conduct genetic balance test;Usingc~2 test to compare genotypes and alleles among groups;the interaction additive model was used to evaluate the relationship between PPAP2B gene-environment interaction and cerebrovascular stenosis.Results:(1)AA/GG genotype and allele frequency of PPAP 2B gene rs 17114036 in cerebrovascular stenosis group were significantly higher than those in control group(OR=1.78,95%CI 1.21~2.66,P=0.003;QR=1.42,95%CI 1.08~2.16,P=0.017);The frequency of TT/CT genotype and T allele at rs 6588635 locus in cerebrovascular stenosis group was significantly higher than that in control group(OR=1.63,95%CI1.14~2.53,P=0.002;OR=1.61,95%CI 1.25~2.70,P=0.001);In addition,the history of hypertension,diabetes,smoking,systolic blood pressure,diastolic blood pressure and blood lipid level in cerebrovascular stenosis group were significantly higher than those in control group(P<0.05).Logistic regression analysis showed that after eliminating confounding risk factors,AA/AG genotype at rs 17114036 and TT/CT genotype at rs 6588635 were still independent risk factors of cerebrovascular stenosis(OR=1.431,95%CI 1.150~2.834,P=0.039;OR=1.713,95%CI 1.221~2.401,P=0.002).(2)The results of transactional analysis showed that there was no interaction between PPAP 2B gene rs 17114036 and rs 6588635 and other environmental risk factors such as body mass index,drinking history,smoking history,hypertension history,diabetes history(P>0.05).Conclusion:(1)PPAP 2B gene rs 17114036 locus A allele may be the susceptibility gene of cerebrovascular stenosis.(2)The T allele at rs 6588635 of PPAP2B gene may be the genetic susceptibility gene of cerebrovascular stenosis.(3)The history of hypertension,diabetes,smoking and hyperlipidemia are risk factors of cerebrovascular stenosis.(4)There was no interaction between PPAP2B gene and the conventional risk factors of cerebrovascular stenosis.
