Background/Aims: Acute myocardial infarction(AMI)is the most prevalent cause of mortality and morbidity worldwide and the number of individuals at risk is increasing.The key to the treatment of this disease is early diagnosis of acute myocardial infarction and early opening of infarct related vessels.In order to further improve the curative effect of acute myocardial infarction and save more people's lives,improved tools for risk prediction including the identification of novel accurate biomarkers are needed.Recent studies have shown that circulating microRNAs(miRNAs)showed significant difference in patients with acute myocardial infarction,and there was a correlation between them.The purpose of this study is to preliminarily study the correlation between miR-17-5p,mi R-126-5p and miR-145-3p and acute myocardial infarction.In order to further study whether miR-17-5p,miR-126-5p and miR-145-3p can become the new biomarkers for acute myocardial infarction,To further improve the specificity,sensibility and timeliness of myocardial injury markers in acute myocardial infarction.Methods: To investigate the kinetic expression of the three selected miRNAs,we enrolled29 patients with AMI and 21 matched controls.Plasma samples were collected from each participant,and total RNA was extracted.Quantitative real-time PCR(q-PCR)was used to investigate the expression of circulating miRNAs.Plasma samples from another age-and gender-matched cohort were collected to compare the expression of the selected miRNAs.Results: Levels of plasma miR-17-5p,miR-126-5p and miR-145-3p were signifcantly increased in early stage of AMI(including before PCI and after PCI).And we found that there no obviously different expression of these miRNA between AMI before PCI and after PCI.Receiver Operating Characteristic(ROC)analysis indicated that each single miRNA showed considerable diagnostic effciency for predicting AMI.Moreover,when combined all three miRNAs,the composited miRNAs presented a higher discriminatory power than any of the miRNAs alone,indicating that the composited miRNAs displayed improved accuracy in identifying AMI.Conclusion: This study was the first to study the difference of plasma circulating miR-17-5p,miR-126-5p and miR-145-3p in acute myocardial infarction before PCI and 1 hours after PCI compared with the control group.And our results show that miR-17-5p,miR-126-5p and miR-145-3p are closely related to acute myocardial infarction.It shows great potential as a new biomarker for early diagnosis of acute myocardial infarction.At the same time,the potential unique characteristics of miR-17-5p,miR-126-5p and mi R-145-3p in patients with acute myocardial infarction have laid a solid foundation for the early diagnosis of acute myocardial infarction and the early treatment of acute myocardial infarction.
