Objective: To explore the effect of crp knockout on metabolism and physiology of rats.Methods:A large number of male wild-type rats and male homozygous rats were obtained in a large number of heterozygous female and heterozygous male rats for mating.All the insulin resistance model rats were started to be fed with high-fat diet at the age of 8 weeks and the modeling time was 12 weeks.The ELISA kit was used to measure the content of insulin in the blood of rats in each group.Western blot technique was used to measure the level of FGF-21 protein in rat liver.Body weight gain,blood pressure heart rate,fasting blood glucose,fasting insulin,energy metabolism,exercise tolerance,and glucose tolerance test(GTT)and insulin tolerance test(ITT)were measured.He staining showed the size of fat cells and the number of hepatic vacuoles.Results: body weight(BW),fasting insulin(Ins),total cholesterol(TC),triglyceride(TG),blood pressure(BP)and respiratory quotient(RER)in wt-hfd rats were significantly higher than those in CRP-HFD rats(p<0.01).Or p<0.05),free fatty acids(FFA),oxygen consumption(VO2),carbon dioxide production(VCO2),were all lower than those in the CRP-HFD group(p<0.01 or p<0.05).The insulin,TC of rats in WT-ND group were higher than those in CRP-ND group(p<0.01 or p<0.05).In the rectal temperature measurement,the rectal temperature in the CRP-HFD group was significantly higher than that in the WT-HFD group(p<0.01),both at room temperature and cold stimulation.In the glucose tolerance test(GTT)and insulin tolerance test(ITT),GTT-AUC,Ins-AUC,and ITT-AUC were smaller in the CRP-HFD group than in the WT-HFD group(p<0.05 or p<0.01),ITT-AUC in CRP-ND rats is smaller compared with WT-ND rats(p<0.05).In the lipid perfusion-hyperinsulin-euglycemic clamp,the glucose infusion rate(GIR)in the CRP-ND group was significantly higher than that in the WT-ND group(p<0.05).In the prolonged hunger test,both the high-fat fed rats and the normal diet-fed rats,CRP gene knockout mice 24 h after fasting blood glucose and liver expression of PEPCK was significantly higher than the wild type rats(p<0.01 or p<0.05).In the exercise tolerance test,exercise tolerance of the crp-nd group was significantly better than that of the WT-ND group(p<0.05).After fat and liver he staining,compared with WT-HFD rats,adipocytes in the CRP-HFD group were significantly smaller,and the hepatic vacuole cells were also significantly smaller.CONCLUSION: After the CRP knockout,the rats' glucose and lipid metabolism are all improved,and they have strong resistance to obesity induced by high fat diet.
