Objectives: By establishing the model of diabetic erectile dysfunction in SD rats,the objective of this study was to observe apoptosis in the cavernous body of the DIED rats and to determine whether there was an increase in apoptosis in the cavernous body of the DIED rats,and to investigate the effects of Schisandrin on apoptosis and the expression of bcl-2?Bax and caspase-3 in the cavernous body of DIED rats,and to explore the effect and the possible mechanism of Schisandrin on erectile function in DIED rats.Methods:1.Set the models SD rats with DIED:Selected 80 healthy 8-week-old male SD rats(body mass 200±50 g),adaptive feed after 1 week,68 rats were randomly injected intraperitoneally with STZ(60 mg/kg),another 12 rats only be regarded as normal control group.After three days,blood glucose was measured by tail vein,the rats with blood glucose ?16.7mmol/L were treated as diabetic rats;8 weeks later DM rats were injected with APO.Those rats who did not have an erection in 30 minutes were seen as DIED rats.2.Group and drug intervention:The rats with DIED were randomly divided into: the DIED group,the low-dose Schisandrin group(5mg/kg/d),and the high-dose Schisandrin group(20mg/kg/d).The NC group was also included.The rats in the NC group and the DIED group were treated with2ml/d of physiological saline,and the drug intervention groups were treated with a corresponding dose of Schisandrin,which lasted for 8 weeks.3.After 8 weeks of gastric lavage,rats were anesthetized and stimulated by electrical stimulation of the penis cavernous nerve and measured the ICP?MAP.4.Decapitate rats and collect cavernous tissue of the cavernous body.TUNEL method was used to detect the apoptosis of cavernous body cells.The expression level of bcl-2,Bax and caspase-3 in cavernous body was determined by Western Blot.Result:1.Changes of blood glucose and body mass in rats before and after gavage:Compared with the NC group,the blood glucose levels before gavage of the rats in the DIED group,the low dose of the Schisandrin group and the high dose of the Schisandrin group were significantly higher than those in the NC group,and the body mass was significantly reduced(P<0.05).In addition to the NC group,there were no significant differences in blood glucose and body mass between the any two groups.(P>0.05).After8 weeks of gavage,compared with the NC group,the blood glucose of the other three groups increased significantly,and the body mass was significantly reduced(P<0.05),and there were no significant differences in blood glucose and body mass between the any two groups.2.There was no significant difference in basic ICP in each group during electric stimulation(P<0.05).There was no significant difference in MAP between the groups in the electrical stimulation(P>0.05).Compared with the NC group,the ICP and ICP/MAP of the DIED group and the low dose of the Schisandrin group and the high dose of the Schisandrin group were significantly reduced(P<0.05).There were no significant differences between the DIED group and low dose of the Schisandrin group(P>0.05).There were significant differences between the DIED group and high dose of the Schisandrin group(P<0.05).3.TUNEL detection results:Compared with the NC group,the apoptosis rate of the cavernous body cells in the DIED group,and the low and high dose groups were all increased(P<0.05).Compared with the DIED group,the apoptosis rate of the cavernous body cells in the low and high dose groups were reduced,and the high dose group was the most obvious(P<0.05).4.Western Blot Result:Expression of protein Bcl-2:Compared with the NC group,the expression of bcl-2 protein in the cavernous body of the other groups was decreasing(P<0.05);Compared with the DIED group,the bcl-2protein in the low and high dose group was higher than that in the DIED group(P<0.05);There were no significant differences in bcl-2 protein expression in low and high dose groups(P>0.05).Expression of protein Bax?Caspase-3:Compared with the NC group,the expression of Bax and Caspase-3 in the cavernous body of the DIED group,low dose of the Schisandrin group and the high dose of the Schisandrin group were all increased(P<0.05);Compared with the DIED group,Bax and Caspase-3protein in the low and high dose groups were lower than that in the DIED group(P<0.05),and the high dose group had the lowest expression(P<0.05).Conclusion:1.The apoptosis rate of the cavernous body cells and the expression of Bcl-2,Bax and Caspase-3 may be involved in the pathogenesis of DIED.Erectile dysfunction may be caused by the increase of apoptosis.2.The high dose of Schisandrin can significantly improve the erectile function of the DIED rats.3.Schisandrin may improve the function of DIED by increasing the expression of Bcl-2 protein,reducing the expression of Bax and Caspase-3protein,and reducing the apoptosis of the cavernous body cells.
