Objective:Objective to study the effects of phthalein components in Angelica Oil on its smooth muscle effect,so as to provide reference for the development of angelica oil.Method:1.Changes of phthalide in Angelica sinensis oil.The content of Z-ligusticide in angelica oil was detected by GC-MS in 5 parts of angelica oil and each 5ml,respectively,under the conditions of-20? centigrade light closure(preservation conditions),25?±5 closed light,25±5no light,40?centigrade light and 40? + 4500 lx.According to the change of Z-ligusticide,we select the condition of angelica oil,take 6 parts of angelica oil,each 10 ml,and examine the content of the components in the angelica oil under the conditions of 0d,15 d,30d,60 d,90d and 120 d under the selected conditions,and analyze the changes of phthalide in the oil under different placing times.2.Effects of angelica oil with different storage time on the movement of smooth muscle of duodenum and uterus in rats.Taking SD rats,the experimental specimens of the isolated smooth muscle of the duodenum and the uterus were taken and fixed in the constant temperature bath filled with nutrient solution.The muscle tension transducer was joined and the angelica oil was added to the different placement time to detect the effect of angelica oil on the movement of duodenum smooth muscle and the smooth muscle of the uterus in rats.3.Correlation between phthalide compounds and smooth muscle effect in Angelica OilUsing the grey correlation analysis method,the change rate of the inhibition rate of rat smooth muscle in different storage time was the parent sequence.The peak area change rate of Z-ligusticide,ligusticide,Z-3-butenyl phthalide and 3-butylene phthalide in the oil of Angelica sinensis at different storage time was subsequence,and the correlation degree was calculated and the oil of Angelica sinensis was analyzed.Effects of changes in phthalide compounds on their smooth muscle effects.Results:1.Changes of phthalide in Angelica sinensis oilThe contents of four phthalphthalein in 4500 lx and 40 C were changed to a certain extent.The content of Z-ligusticide decreased from 59.64% to 47.23%,ligusticide decreased from 5.93% to 3.14%,and the content of Z-3-butylene phthalide increased from 2.11% to 3.05%,and the percentage of 3-butylene phthalide increased from8.47% to 15.53%.Conclusion: the four main phthalates of angelica oil have certain instability.There may be some transformation relationship between phthalein substances in angelica oil.2.Effects of angelica oil with different storage time on the movement of smooth muscle of duodenum and uterus in rats.By pre experiment,we found that angelica oil could inhibit the spontaneous contraction of duodenum in vitro,and put it in the accelerated condition 120 d,0.01%angelica oil showed a decreasing trend,and the inhibition rate dropped to 27.62%.Through the pre experiment,we found that angelica oil showed a two-way effect on the isolated rat uterine smooth muscle.Small dose(0.01%)was excited,high dose inhibition,it was placed in the accelerated condition 120 d,0.1% angelica oil inhibited the inhibitory rate of uterine smooth muscle from 45.41% to 16.35%.3.Correlation between phthalide compounds and smooth muscle effect in Angelica OilThe changes of Z-Ligusticum,ligusticide,Z-3-butenyl phthalide and 3-butylene phthalphthalide in angelica oil are related to the effect of angelica oil smooth muscle,and the weight of the influence is Z-Ligusticum > Ligusticum > 3-butylene phenolphthalide > Z-3-butenyl phthalide.Conclusion:The Z-ligusticide and other phenolphthales in angelica oil can be degraded or converted under certain conditions.This change will have an important effect on the smooth muscle effect of angelica oil;the development and utilization of the smooth muscle effect of angelica oil should pay attention to the stability conditions of phthalphthalein.
