The Expression And Significance Of MMP-2 And HMGB-2 In Preeclampsia

Objective: Exploring the expression and significance of MMP-2 and HMGB-1 in maternal serum and placenta tissue in preeclampsia(PE).Discussing the correlation with PE,and analyzing the possible mechanism and clinical applications.Methods: 37 cases of the pregnants with PE in Gansu Provincial Hospital from December,2016 to August,2017 were selected as the group PE,and 40 cases of the normal pregnants were divided into group control.To navigate the qualitative expression of the HMGB-1,MMP-2 protein in placenta tissue using Immunohistochemical staining;ELISA had been used to detect the expression of MMP-2 and HMGB-1 in maternal serum within the pregnancy,and western blot had been used to detecte the expression of MMP-2 and HMGB-1 in placenta tissue after childbirth.The results: 1.Immunohistochemistry results 1.1 Immunohistochemistry showed that the expression of HMGB-1 group of normalpregnancy were mainly located in placental syncytiotrophoblasts,in vascular endothelialcells and a few in the stromal cells of cell nucleus.The expression of HMGB-1 inpreeclampsia group placenta were mainly located in placental syncytiotrophoblasts invascular endothelial cells and a few in the stromal cells of cell nucleus and cytoplasmic.The expression of MMP-2 group of normal pregnancy were mainly located in villussyncytiotrophoblast cells,cytotrophoblast cells and trophoblastic cells outside the villi,placental villi of vascular endothelial cells,interstitial cells and a part of decidual cells.The expression of MMP-2 in preeclampsia group placenta were reduced,or evencompletely lost in the above parts.1.2 The expression of MMP-2 in placenta of preeclampsia group was lower than that of thecontrol group(P <0.05),the expression of HMGB-1 was higher than that of the controlgroup(P <0.05).The differences in each group were statistically significant.2.ELISA assay results showed The serum level of MMP-2 in preeclampsia group was lower than that in control group(P <0.05).The serum HMGB-1 level in preeclampsia group was higher than that in control group(P <0.05).The differences between the two groups were statistically significant.3.Western blotting results showed The results of Western blotting showed that the expression of MMP-2 in placenta of preeclampsia group was significantly lower than that in control group(P <0.001),and the expression of HMGB-1 was significantly higher than that in control group(P <0.001).Conclusion: The expression of MMP-2 and HMGB-1 were closely related to the occurrence of PE.A low of MMP-2 and over-expression of HMGB-1 showed that it could be involved in the pathogenesis of PE.
