The Changes And Significance Of BAFF And IL-10 In Adjuvant Treatment Of Children With SLE With Huaiqihuang Granules

Objective:By observing the changes of SLEDAI scores,laboratory parameters,serum BAFF,and IL-10 in children with systemic lupus erythematosus(SLE),we investigated the efficacy and safety of Huaiqihuang granules(HQH)in the adjuvant treatment of children with SLE.Methods:Thirty-four children with SLE who were admitted to Shengjing Hospital Affiliated to China Medical University were selected as the study subjects.The time of case collection was 2016.1-2017.6.The patients were divided into groups A and B according to the randomized control method.Treatment group,a total of 18 cases,2males and 16 females,the average age of 10.2 ± 2.36 years old,given glucocorticoid and(or)immunosuppressive agents and Huaiqihuang granules orally;B group for the conventional treatment group,a total of 16 Cases,1 male and 15 female,mean age9.73±2.49 years,given glucocorticoids and(or)immunosuppressive therapy;10 cases of children in the same period of childhood health examination physical examination as the normal control group(group C There were 1 male and 9 female subjects with an average age of 8.5±2.46 years.There was no significant difference in age and sex between the three groups(p<0.05).Serum levels of BAFF and IL-10 were measured by ELISA.Treatment was observed at the end of March to compare SLEDAI scores and laboratory indicators.Result:(1)SLEDAI score: 11 cases in group A(5-9 points),4 cases(10-14 points),3cases(?15 points);9 cases in group B(5-9 points),4 cases(10-14 points),(?15 points)2 cases,(<5 points)1 case;before treatment,there was no significant difference between the two groups SLEDAI scores(p> 0.05);after treatment,both groups SLEDAI scores decreased(p <0.05),group A The decrease was greater than that in group B(p<0.05);(2)Serum BAFF,IL-10 levels: Before treatment,serum BAFF and IL-10 levels were higher in group AB than in group C,and there was a significant difference between groups(p<0.05).There was no difference in AB group(p>0.05).After treatment,serum BAFF and IL-10 levels in both groups were decreased(p<0.05).The decrease in group A was greater than that in group B(p<0.05).Correlation analysis showed that serum IL-10 levels were positively correlated with SLEDAI scores(p<0.05),serum BAFF levels were not correlated with SLEDAI scores(p>0.05);(3)Immunoglobulins: Before treatment,both groups had higher than normal range of immunoglobulins,with no comparison between groups.Significant difference(p>0.05);after treatment,Ig G decreased significantly(p<0.05),group A decreased more than group B(p<0.05),Ig A,Ig M did not change significantly(p>0.05);(4)complement: before treatment Compensatory reduction in two groups,comparison between groups Significant differences(p>0.05);After treatment,the two groups of complements increased(p<0.05),the increase in group A was greater than in group B(p<0.05);(5)Lymphocyte subsets: two groups of CD3+,CD4+,and CD4+ before treatment.The proportion of CD8+ and NK cells was low,and the proportion of CD8+ and CD19+ was high.There was no difference between the groups(p>0.05).After treatment,the proportion of CD19+ decreased(p<0.05),the proportion of CD3+ increased(p<0.05),and increased.Compared with the reduction rate,group A was greater than group B(p<0.05);there was no difference in CD8+,CD4+,CD4+/CD8+,and NK cells before and after treatment(p>0.05);(6)autoantibodies: 1ANA titer: before treatment,two The ANA titers were elevated in all groups.After treatment,the rate of decline in group A was greater than that in group B(100%,73.3%);2ds-DNA antibodies: Before treatment,both groups were positive for ds-DNA antibodies.After treatment,group A and group 2 Turned negative,no one in group B turned negative.(7)24-hour urinary protein: Before treatment,7 patients in group A(? 25 mg/kg),9patients(25-50 mg/kg),2 patients(?50 mg/kg),and 7 patients in group B(?25 mg/kg).(25-50 mg/kg)in 4 cases(?50 mg/kg)in 5 cases.The 24-hour urinary protein decreased in the two groups after treatment.There was no difference between the two groups(p>0.05).Conclusion:In the treatment of children with SLE,Huaiqihuang Granules may reduce serum BAFF levels,reduce B cell activation,decrease IL-10 secretion,regulate T and B cell imbalance,reduce autoantibody production,and promote disease recovery.
