Objective: To detect the expression of PGRMC1 in amniotic membrane and chorionic villi of PPROM patients by Western-Blot and Real-Time PCR,and to explore its clinical significance.METHODS: Western-Blot and Real-Time PCR were used to detect the expression of PGRMC1 in 10 cases of preterm premature rupture of membranes and in 10 cases of cesarean section without ectopic contractions at the same time.The real-time PCR method was used to detect the expression of IL-1β and TNF-α in the amniotic membrane and chorion of preterm premature rupture of membranes.RESULTS: The expression of PGRMC1 was mainly located in the chorion and it was also expressed in the amnion.The expression level of PGRMC1 in the chorionic tissue of preterm premature rupture of membranes was significantly lower than that of the control group(P<0.05).Compared with the control group,the expression of IL-1β and TNF-α in the amniotic membrane and chorionic villi was increased in the PPROM group(P<0.05).The expression level of PGRMC1 was correlated with the expression level of IL-1β(R=-0.64,P<0.05).Conclusion: The expression level of PGRMC1 in fetal membranes of premature rupture of membranes is significantly lower than that of control group.The expression of PGRMC1 is correlated with the expression level of IL-1β in inflammatory mediators.PGRMC1 may be used as a method to treat preterm premature rupture of membranes.Target.
