Pinellia Rhizoma is the dry tuber of the Pinellia ternata(Thunb.)Breit.,which has the functions of drying dampness and dispersing phlegm,antiemetic effects,reducing swelling and resolving mass.It is commonly used to treat dampness,cold sputum,cough,phlegm,phlegm,dizziness,wind phlegm,headache,vomiting and nausea.The Department of Health take Pinellia Rhizoma as one of the 28 kinds of toxic traditional Chinese Medicine?When used clinically,because of its toxicity,it is often used as medicines after processing to attenuate the drug.The traditional processing methods of Pinellia Rhizoma are often added to accessories(ginger,alum,limewater)in order to reduce toxicity and increase efficacy.However,the processing technology is complex,the processing time is longer,and processing involves long time soaking or decocting,which can cause the loss of effective components.Therefore,it is necessary to explore a new process that can replace the traditional one.Present study shows that: A high-pressure steaming method can reduce the taste and irritation of Pinellia Rhizoma,shorten processing time and improve production efficiency.However,the principle of high pressure evaporation and the principle of reducing irritation were not studied thoroughly.Based on this situation,this subject proposes to use a high-pressure steaming method to process Pinellia Rhizoma.Firstly,the changes in toxicity,irritation,and pharmacodynamics of Pinellia Rhizoma after the high-pressure steaming process were preliminarily studied to ensure its safety and efficacy.Secondly,the response surface methodology was used to optimize the best processing parameters in order to provide reliable basis for the establishment of new technology of Pinellia Rhizoma high pressure steaming.Finally,the main chemical constituents of Pinellia Rhizoma were measured before and after processing in order to find out the changes,and to provide certain scientific basis for the analysis of the effects of processing on reducing toxicity and reservating /increaseing effect.The main research contents and results are as follows.For the toxicity and efficacy,the acute toxicity test of mice and conjunctival irritation test of rabbit eyes were used to evaluate the toxicity and irritation of Pinellia Rhizoma before and after steaming.By establishing an allergic asthma model and assessing the functions of drying dampness and dispersing phlegm of Pinellia Rhizoma were evaluated before and after the steaming.The experimental results showed that the toxicity of steamed Pinellia Rhizoma was lower than that of unprocessed Pinellia Rhizoma.Meanwhile,Steamed Pinellia Rhizoma is mildly stimulus,and its irritative effect is significantly lower than that of unprocessed Pinellia Rhizoma.Both Pinellia Rhizoma and steamed Pinellia Rhizoma have certain effects on allergic asthma in rats.The effect of steamed Pinellia Rhizoma on the pathological status of lung tissue in rats is better than that in Pinellia Rhizoma.At the same time,there is no significant difference of the number of EOS cells,the content of IL-4,IFN-? in BALF and IgE in serum by the steamed Pinellia Rhizoma and the Pinellia Rhizoma.For the processing technology,based on the single factor experiment,the subject adopts response surface method based on Box-Behnken design.The optimum processing technology for high pressure steaming of Pinellia Rhizoma is selected,with infiltration time,steaming temperature and steaming time as the inspection factors,comprehensive evaluation scores of leachables and organic acids as evaluation indexes.The optimum steaming process parameters were: infiltration time 9.5 h,steaming temperature 118 ?,and steaming time 2.0 h.For the chemical composition in this subject,the structure and morphology of calcium oxalate crystals were observed by microscope and electron microscope.At the same time,the changes in content of the chemical constituents of Pinellia Rhizoma,like organic acids,alkaloids,polysaccharides and calcium oxalate crystals were determined before and after the processing by ultraviolet spectrophotometer,potentiometric titrator,and high performance liquid chromatography.The changes in chemical composition before and after processing were analyzed to clarify the effect of steaming on the chemical composition of Pinellia Rhizoma,in order to further explain the rationality of reducing toxicity and reservating /increaseing effect of high-pressure steaming in terms of the chemical composition.The experimental results showed that after steaming,a variety of chemical components of Pinellia Rhizoma have undergone significant changes in varied degrees in chemical composition.The morphology of calcium oxalate crystallites changed significantly after steamed with small change of content,the content of alkaloids decreasing,the content of organic acids and polysaccharide increasing significantly?To sum up,the subject carried out the research of high-pressure steaming of Pinellia Rhizoma processing technology,studied the toxic effects and chemical composition changes before and after the processing of Pinellia Rhizoma,and confirmed that high-pressure steaming can achieve the effect of reducing toxicity and reservating /increaseing effect.At the same time,the best process of Pinellia Rhizoma steaming is established,which provides a reliable basis for the establishment of new technology of high pressure steamed Pinellia Rhizoma,and also provides a new idea for enriching and developing the kinds of Pinellia Rhizoma artillery products.
