Psoralen,bitterness and dryness,can warmly invigorating kidney yang,warm spleen and antidiarrheal,can connect the fire of the life gate and pericardium,it has therapeutic effects on primordial YANG languishing,Kidney deficiency and low back pain inSolid spirit,Diarrhea with spleen deficiency and so on.At present,there are different degrees of research on its processing of metaria medica technology and processing mechanism.The theory of"Lei Gong Pao Zhi Lun"describes it as"Sexual dryness poison".In the Compendium of Materia Medica,there are records of"salt water immersion and stir fried into blood and moistening dryness."Therefore,this study combines traditional Chinese medicine theory into"Salt into the kidney","salty moistening"and so on.A preliminary study on the Fructus Psoraleae with salt processing slow down the dryness poison was made.It is hoped that it will provide a more detailed and sufficient theoretical basis for the clinical medication guidance of the drug.ObjectiveChoosen a single Chinese medicine psoralen as the research material of this topic,The change of dryness of Fructus psoralen after salt processing was studied.Adopt the research mode of"pharmacology research-metabolism analysis-molecular biology research",embody synergism and moisturizing dryness syndrome through pharmacological effects.Spectrum effect combination,cocktail probe method and fluorescence quantitative PCR technology were used.Combining model animal with TCM syndrome type,based on the pharmacological effects,this paper analyzes the processing principle of psoralen salt drying and its dryness in a deep level to the gene level.In order to deepen the understanding of processing theory and the development of Chinese medicine processing discipline,add bricks and mortar to it.MethodAccording to the process parameters established by the previous research results of the project group,the preparation of psoralen and salt broiled pieces was prepared,and the samples were prepared according to the corresponding extraction conditions-alcohol water double extraction method.The model animals were replicated by hydrocortisone and the effects of the related indexes on the normal and model animals before and after the processing of psoralen were selected.To evaluate the effects of psoralen salt on liver and kidney function,liver enzyme subtypes and aquaporins.1.Liver and kidney function indexes were used as indicators:AST,ALT,TP,Alb,Scr and BUN.To investigate the effects of psoralen salt on the function of liver and kidney in rats with kidney yang deficiency.2.The effect of caffeine CYP1A2,CYP2E1,CYP3A4 as a probe,and antipyrine as the internal standard drug,to explore the effects of psoralen and salt on the activity of the 3 major metabolic enzyme subtypes,CYP1A2,CYP2E1 and CYP3A4,of the CYP450 enzyme system.3.With the change of water metabolism as the main object,the expression of AQP5,AQP4 and AQP2 mRNA in salivary glands of oral cavity were measured by fluorescence quantitative PCR?qRT-PCR?method.The effects of different processed products on the regulation and expression of AQP5,AQP4 and AQP2 were studied,and the principle of salt drying to relieve dryness was analyzed.Result1.Compared with the model group,ALT and AST in the serum of the raw Fructus Psoralea and the Fructus Psoraleae with salt processing group increased significantly?P<0.01?,and Alb decreased significantly?P<0.01?.The levels of ALT and AST in the serum of the Fructus Psoraleae with salt processing group were lower than those in the raw Fructus Psoralea group,and the Alb level was higher in the psoralen group than in the raw products group.Compared with the control group,the Scr of the rats in the model group decreased significantly?P<0.05?,and the BUN did not change significantly.The levels of BUN and Scr in each drug delivery group increased significantly compared with the model group,with a statistically significant difference?P<0.05?.2.Effect on t1/2:the caffeine t1/2 in the raw product group was significantly higher than that in the control group?P<0.05?.The t1/2 in the raw and salt broiled group was significantly lower than that of the control group?P<0.05?.The caffeine,the sulfoxide and the chlor t1/2 were significantly lower than the raw group?P<0.05?.The influence of the apparent volume V:the caffeine in the salt group and the V of the sulfoxide were significantly smaller than the control group and the raw product group?P<0.05?.The V of the raw and salt broiled group was significantly smaller than that of the control group.The effect of CL on the clearance rate was significantly higher in the salt group than in the control group?P<0.05?.The CL in the salt group was significantly higher than that in the control group?P<0.05?.he effect on the area AUC0-infinity under the curve:the AUC0-infinity of the raw product group and the salt broiling group was significantly lower than that of the control group?P<0.05?.And the AUC0-of the chloride group in the salt group was significantly lower than that of the the control group?P<0.05?.3.The expression of AQP2 and AQP4mRNA in model group was significantly lower than that in control group?P<0.05?,while AQP5mRNA increased significantly?P<0.05?.The expression of AQP4mRNA in the Fructus Psoraleae with salt processing group increased significantly compared with the model group?P<0.05?,and the expression of AQP5mRNA decreased significantly?P<0.05?.Compared with the Fructus Psoraleae with salt processing group,the expression of AQP2 and AQP5mRNA in Stir fried with the Fructus Psoraleae with salt processing group increased significantly?P<0.05?,and AQP4mRNA expression was significantly decreased?P<0.05?.ConclusionThe raw Fructus Psoraleae has certain damage to the liver function of rats,which may be relieved with salt processing.After salt processing,psoralen can reduce the inhibition of CYP1A2 and enhance the induction of CYP3A4 and CYP2E1,that is,the metabolism of CYP1A2,CYP3A4 and CYP2E1 metabolites after psoralen is increased,the metabolic rate of the metabolites is increased,the concentration of blood drugs is reduced,and the dryness of psoralen is alleviated.Psoralen with salt processing can relieve the dryness,may be related to the regulation of gene expression of AQP in the body.Compared with psoralen,psoralen can increase the expression of AQP2 and AQP5mRNA,reduce the expression of AQP4mRNA,and suggest that the dryness of drugs may be relieved after the salt broiling.
