Fructus Psoraleae is a common Chinese traditional medicine in clinic. The edition of Pharmacopeia in 2015 contains two varieties-Raw Fructus Psoraleae and salt-processed Fructus Psoraleae.The raw Fructus Psoraleae has strong actions of warming the kidney and invigorating spleen, but The raw Fructus Psoraleae can cause some symptoms of impairment of yin such as dry mouth?sore throat because of its property "warm" and its flavor "pungent" and its dryness if used for long time or in over-dose. The Fructus Psoraleae after salt processing has a better action on warming kidney and invigorating spleen by moderating its property "pungent,dry",so the Fructus Psoraleae after salt processing is used more widely in clinic. At present, some research have studied the processing technology, the changes of chemical composition and pharmacological effects before and after processing, while there is little research about that the salt-processed Fructus Psoraleae has moderated dryness and toxicity. Thus, this research studied the relevant problems about the moderated dryness and toxicity of salt-processed product.ObjectiveOur study aimed at traditional medicinal herb Fructus Psoraleae, focused on moderated dryness and toxicity of Fructus Psoraleae with salt processing, proceeded in these aspects such as chemical components, the mechanism of pharmacology, spectrum-effect relationship, expecting to elucidate active components in salt-processed Fructus Psoraleae for moistening dryness and toxicity.MethodThe two processed products-Saw Fructus Psoraleae and Fructus Psoraleae with salt processing in the study were prepared according to the process parameters worked out by previous study and the test articles are prepared by reference to the extraction conditions of previous study. With establishing an animal model of deficiency in kidney-yang caused by hydrocortisone and another animal model of diarrhea induced by Rheum officinale causing spleen weakness, taking the actions of reinforcing kidney and moderating dryness and toxicity as evaluation indicators, the study researched on the synergy and moderating dryness and toxicity effects of salt-processed Fructus Psoraleae, the relationship between of its the synergy and moderating dryness and toxicity effects and its processing technology, and then to clarify the active components in salt-processed Fructus Psoraleae for moistening dryness and toxicity in these aspects such as chemical components, the mechanism of pharmacology, spectrum-effect relationship.1. Taking the indexes about organs, content of cAMP/cGMP, TNF-a,Na+-K+-ATP in serum as evaluation indicators, the study observed the different dryness and toxicity of Raw Fructus Psoraleae and its salt-processed product in different doses acting on the rats with deficiency in kidney-yang induced by hydrocortisone and the rats with diarrhea caused by spleen weakness separately.2. Taking the indexes about organs, content of cAMP/cGMP,TNF-a,Na+-K+-ATP in serum as evaluation indicators, the study observed the different actions of different processed products of Fructus Psoraleae-raw products, salt-processed products, water-processed products on the rats with deficiency in kidney-yang induced by hydrocortisone and the rats with diarrhea caused by spleen weakness separately.3. Taking the indexes about organs, content of cAMP/cGMP, TNF-a,Na+-K+-ATP in serum as evaluation indicators, the study observed the different actions of the salt-processed product's extract in different solvents-petroleum ether, ethyl acetate, n-butyl alcohol on the rats with deficiency in kidney-yang induced by hydrocortisone.4. The study established the HPLC finger print of the Raw Fructus Psoraleae's ethyl acetate extract, established an animal model of deficiency in kidney-yang by injecting hydrocortisone into rats, and gave the rats the ethyl acetate extract of Fructus Psoraleae by gavages, analyzed the correlation of common peaks'area in HPLC finger print and dryness and toxicity indexes with grey correlation degree analysis method, expecting to clarify the active chemical components of inducing dryness and toxicity in Raw Fructus Psoraleae in ethyl acetate extract.Result1. High dose Raw Fructus Psoraleae (7k/kg) had significant increased actions on rats'liver and kidney index,cAMP/cGMP,Na+-K+-ATP and lower actions on rats' thymus index(p<0.05).2. Raw Fructus Psoraleae had significant increased actions on rats'liver and kidney index,cAMP/cGMP,Na+-K+-ATP and lower actions on rats'thymus index(p<0.05).No significant effect of water after processing by, there are significant differences and after salt processing (p<0.05).3. The petroleum ether extract of Raw Fructus Psoraleae had significant increased actions on kidney index,cAMP/cGMP,TNF-a content of the animal model with kidney yang deficiency(p<0.05);The ethyl acetate extract of Raw Fructus Psoraleae had significant increased actions on liver and kidney index,TP,GPT,BUN content and lower actions on thymus index and Alb of it(p<0.05);While the n-butyl alcohol extract of Raw Fructus Psoraleae had significant increased actions on kidney index,TP,cAMP/cGMP,and Na+-K+-ATP content and lower actions on Alb and TNF-a content of it(p<0.05);In addition, the water extract had significant increased actions on liver and kidney index and lower actions on thymus index, Alb and TNF-a content of it(p<0.05).4. These common peaks of ethyl acetate extract (ID) No.2,3,11,12, 14,16,17,18 had high degree of associations with liver function while other common peaks (ID) No.2,3,5,6,14,22 had high degree of associations with kidney function. In these common peaks, the peaks No.14 and the peaks of psoralen, isopsoralen all had high degree of associations with toxic dryness and toxicity.ConclusionThe dryness and toxicity of Fructus Psoraleae could increase with increasing dose. The salt-addition and heating processing technology of salt-processed product could moderate its raw product's dryness and toxicity, dryness and toxicity ingredients mainly exist in ethyl acetate extract and n-butyl alcohol extract of Raw Fructus Psoraleae, and dryness and toxicity of Raw Fructus Psoraleae is caused by the cooperation of a few components in Raw Fructus Psoraleae while the moderated dryness and toxicity may be related with the decreasing of relevant components.
