Objiectiv:through unilateral ureteral obstruction(UUO)to make renal fibrosis model,to study the effect and mechanism of MMP-9,TIPM-1 and HGF in the development of renal fibrosis.To study the influences of different doses of Kangfuxin liquid on UUO renal fibrosis model rats' renal tissue's MMP-9 and TIMP-1 and HGF,observe the impacts of Kangfuxin liquid on renal fibrosis model rats' pathology and renal function,which can proivde a new idea to the treatment of renal fibrosis.Methods:The 108 SD rats were randomly divided into 6 groups: sham operation group,model group,Kangfuxin liquid low/medium/high dose group,and benapuli group.Each group of 18 rats was divided into 7d,14 d and 21 d groups.The renal fibrosis model was established by UUO in addition to the sham operation group.The rats in the sham group had to separate the ureter and not ligate.Building the next day after the rats began to fill the stomach,Kangfuxin liquid low/medium/high dose group were given low/medium/high doses of new liquid recovery,the captopril group to shell the split,control group and model group the same amount of saline lavage respectively.The rats were killed in batches after 7 days,14 days and 21 days,while the serum was produced and pathological specimens were produced from the left obstructive kidney tissue.Biochemical detection serum Scr,BUN,beta 2-MG,CysC;HE staining and Masson staining observed the pathological changes of renal tissue in rats.Immunohistochemical staining was used to observe the expression of HGF,MMP-9 and TIMP-1 in renal tissues,and statistical analysis was performed with SPSS20.0.Results:(1)pathological changes of renal tissue: no obvious pathological changes were observed in the sham operation group.The model group and the treatment group showed different levels of inflammatory cell infiltration,renal tubular atrophy and renal interstitial fibrosis.Among them,the model group and the low dose of Kangfuxin liquid were heavy and the fibrosis lesion was heavier.The model group and treatment group increased the degree of fibrosis over time.The indexes of renal function were: compared with the other groups of Scr,BUN,and ?2-MG and CysC in the sham operation group at the same time period,the content of the sham operation group was the lowest,and the difference was statistically significant(P < 0.05).At the same time,compared with the model group,the content of Scr,BUN,?2-MG and CysC in the model group was the highest,with the difference being statistically significant(P < 0.05).(3)the kidney tissues immunohistochemical results: 1.The MMP-9 in the renal tissue expression: compared with time control and the rest of the group,control the highest expression of MMP-9,the difference was statistically significant(P < 0.05);The mmp-9 expression of the model group was the lowest in the model group compared with that of the high-dose group and the benazole group,and the difference was statistically significant(P < 0.05).2.Timp-1 was expressed in renal tissue: at the same time,compared with the treatment group and the model group(P < 0.05),the expression of timp-1 was lowest in the sham operation group.At the same time,the model group was compared with the high-dose group and the benazril group(P<0.05).3.Expression of HGF in renal tissue: compared with the model group and the treatment group,the expression of the sham operation group was the lowest in 21 days,and the difference was statistically significant(P<0.05).The model group had the lowest expression of HGF in the model group compared with the high dose and the benazupril group in the new fluid in the 21 days,and the difference was statistically significant(P < 0.05).Conclusion:(1)unilateral ureteral obstruction can establish a rat model of renal fibrosis.(2)Kangfuxin liquid can reduce the inflammatory cell infiltration and fibrous tissue deposition in rats with renal fibrosis,suggesting that the Kangfuxin liquid has a certain anti-renal fibrosis effect.Kangfuxin liquid can reduce the content of Scr,BUN,?2-MG and CysC in serum,suggesting that the Kangfuxin liquid has certain protective effects on renal function.(3)Kangfuxin liquid resistance renal fibrosis may be by promoting rat kidney HGF expression,thereby directly increase the expression of MMP-9 in the kidney tissues,and HGF reduction of MMP-9 inhibitor TIMP-1 expressed in rat kidney tissues,indirectly increase the activity of MMP-9,promote the expression of MMP-9,increase the degradation of the ECM.
