Objective:We determined the glucose metabolism,lipid metabolism,bone metabolism and bone mineral density(bone mineral density,BMD)in postmenopausal women in Shihezi,Xinjiang.At the same time,we measured the polymorphism of Wnt3 a and discussed its correlation.To provide the basis for the study of the pathogenesis and target therapy of type 2 diabetes(type 2diabetic Mellium,T2DM)and abnormality of bone metabolismand.Methods:1.The postmenopausal women were selected as the research object in Shihezi community.Recording to the history of diabetes or glucose tolerance test and bone mineral density.They were divided into four groups.26 cases was collected in normal glucose tolerance and normal bone mass group,regard as control group A.28 cases was collected in normal glucose tolerance and abnormal bone mass group,regard as group B.27 cases was collected in type 2diabetes with normal bone mass group,regard as group C.55 cases was collected in type 2diabetes with abnormal bone mass group,regard as group D,amount to 136 cases.2.General clinical information including age,menopausal years,height,weight,body mass index(BMI)and waist-hip ratio(WHR)were measured and calculated.Fasting plasma glucose(FPG),triglyceride(TG),high-density lipoprotein(HDL-C),low density lipoprotein(LDL-C),blood calcium,blood phosphorus and alkaline phosphatase(ALP)were determinant by automatic biochemical analyzer.Hemoglobin a1c(Hb A1c)was measured by high pressure liquid chromatography.Bone density of lumbar1-4 and femoral neck were measured by dual energy X-ray machine.3.Extracted 5 milliliter venous blood and picked up the DNA.4.The polymorphism of Wnt3 a gene(rs752107,rs708114)checked by time of flight mass spectrometry.Results:1.The comparison of general data and biochemical indexes among groups:compared with A group,the age(69.71 + 7.26,70.44 + 6.44)and menopausal years(19.71 + 7.26,20.44 + 6.44)in B?D group showed a high level,which indicated that the baseline date were incomparable.Analyzed the biochemical indexes and bone mineral density after removing the influence of age and menopausal years with covariance analysis.Compared with A group,the FPG(5.18±2.13;7.60±2.13,7.50±2.13)and Hb A1c(5.92±1.06;7.19±1.05,7.50±1.05)in C?D group showed a high level(P<0.01).The TG(2.50±1.52;1.51±1.50,1.70±1.52)?L1-4BMD(1.189±0.15;0.845±0.14,0.911±0.15)and F-BMD(0.882±0.18;0.715±0.19,0.754±0.19)in B?D group showed a low level.(P<0.05).2.The comparison of genotype and allelic genes frequency of rs752107: compare with group A,there were no significant differences in the distribution of rs752107 genotype and allelic genes in group B,group C and group D.(P=0.37,P=0.87,P=0.65);(P=0.49,P=0.80,P=0.38)3.Gene polymorphism of rs752107:There was no significant difference in the relationship between type 2 diabetes and additive genetic models,explicit models and recessive models(P=0.656,P=0.542;P=0.821;P=0.355).The relationship with bone mass abnormality showed no statistically significant difference(P=0.217,P=0.154;P=0.903;P=0.177).4.The comparison of genotype and allelic genes frequency of rs708114:compare with group A,there were no significant differences in the distribution of rs08114 genotype and allelic genes in group B,group C and group D.(P=0.21,P=0.70,P=0.16);(P=0.15,P=0.49,P=0.12)5.Gene polymorphism of rs708114:There was no significant difference in the relationship between type 2 diabetes and additive genetic models,explicit models and recessive models(P=0.395,P=0.471;P=0.611;P=0.410).The relationship with bone mass abnormality showed no statistically significant difference(P=0.897,P=0.372;P=0.185;P=0.790).6.The comparison of two loci polymorphism in each group of different genotypes and biochemical indexes:in A group the HDL-C level of TT/TC carriers was higher than that of the CC type(P=0.13).In C group,the TG level of the TT/TC carriers was lower than that of the CC type(P=0.04)in the rs752107 locus polymorphism;In the A group,the fasting plasma glucose and femoral neck bone density level of CC/CT carriers was lower than that in TT type(P=0.033,P=0.022).In the C group,the fasting plasma glucose and glycosylated hemoglobin level of CC/CT carriers was lower than that of TT type(P=0.000,P=0.005).In the D group,the triglyceride level of CC/CT carriers was higher than that of TT(P=0.041)type in the rs70814 locus polymorphism.Conclusion:1.There were no significant differences in genotype frequency and allele frequency dist ribution of rs752107 and rs708114 loci in different bone metabolic states in 136 case s of postmenopausal women in Shihezi,Xinjiang.2.The polymorphism of Wnt3 a rs752107 locus in this population may have an effect on the level of HDL-C and TG,and the polymorphism of rs708114 locus may affect the level of FPG,Hb A1 c,TG and F-BMD.
