Abstract/Summary:
Objective:The expression level of Dkk1 protein and the level of Dkk1,β-catenin gene were tested in peripheral blood leukocytes,bone mineral density(BMD)was measured in postmenopausal women patients with type 2 diabetes mellitus.To investigate and analyze the relationship between Wnt signaling pathway and bone metabolism in postmenopausal women with T2 DM.Method: 1.50 cases was collected in NGT group and T2 DM group respectively from community and hospital of Shihezi area in Xinjiang during October 2015 to October 2016,general clinical information including sex,age,height,weight,systolic blood pressure(SBP),diastolic blood pressure(DBP)were measured and recorded,body mass index(BMI)was calculated according to the BMI formula.Fasting blood glucose was collected in tube anticoagulated with EDTA,Fasting Plasma Glucose(FPG),serum cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C)and other biochemical indicators were measured by automatic biochemical analyzer,glycosylated hemoglobin was measured by high pressure liquid chromatography;bone density of lumbar and femoral were measured by dual energy X-ray(DEXA)machine.2.The expression level of Dkk1 protein in serum was determined by enzyme-linked immunosorbent assay(ELISA).3.The expression level of Dkk1 and β-catenin gene was detected by fluorescent quantitative reverse transcription polymerase chain reaction(qRT-PCR).4.Data was calculated by SPSS17.0 statistical software.The data were expressed as mean ± standard deviation,and the mean values of the two groups were compared by t test.Non-normal data was tested by nonparametric test.The correlation between variables was analyzed by multiple linear regression,P<0.05 revels that the difference was statistically significant.Results:1.There was no significant difference of age,SBP,DBP,and BMI between the NGT group and the T2 DM group(P<0.05),which indicated that the baseline data were homogeneous and comparable.2.Compared with NGT group,the level of carbohydrate metabolism index such as Fasting Plasma Glucose(FPG)(5.47±0.73,9.26±2.38),(Froctosamine,FMN)(239.74±36.54,337.66±87.31),HbA1c(6.23±0.92,8.70±1.76)(P<0.01)in T2 DM group showed a higher level;There were no significant difference of calcium(Ca)and alkaline phosphatase(ALP)in two groups,while the bone metabolism index such as phosphorus(P)(1.11±0.12,1.24±0.16)showed a higher level,osteocalcin(OC)(30.25±7.41,19.27±5.07),25(OH)VitD3(35.37±6.83,22.00±5.26)(P<0.05)showed a lower level in Postmenopausal women with T2 DM.3.Compared with NGT group,the level of Fins(8.20±2.72,9.29±2.57)and HOMA-IR(2.00±0.70,3.75±1.14)was higher and the level of Fincp(0.48±0.10,0.30±0.09)were lower,HOMA-β and ISI were lower in T2 DM group(P<0.05).4.Compared with NGT group,the level of bone mineral density(BMD)in postmenopausal women with T2 DM showed a lower level of femur(P=0.003).5.Compared with NGT group,the expression of Dkk1 protein showed a higher level in Postmenopausal women with T2DM(209.37±32.35,232.97±39.31)(P<0.05).6.The expression level of Dkk1 gene showed 5.99 times in T2 DM group than NGT group,while β-catenin showed a lower level with 0.46 times in T2 DM group.7.The correlation analysis showed that BMD(L2)were negative related to FMN(r=-0.204;p=0.042),BMD(L3)was positive with 25(OH)VitD3(r=0.201;p=0.045),BMD(Femur)was negative related to FBG(r=-0.344;p=0.000),BMD(Femur)was positive related to Fincp(r=0.244;p=0.025)and HOMA-β(r=0.298;p=0.003);OC showed negative relationship with FPG(r=-0.527;p=0.000),FMN(r=-0.450;p=0.000),HbA1c(r=-0.459;p=0.000)and HOMA-IR(r=-0.423;p=0.000),while positive related to Fincp(r=0.558;p=0.000),ISI(r=0.376;p=0.000),HOMA-β(r=0.419;p=0.000);25(OH)VitD3 was negative related to FPG(r=-0.573;p=0.000),FMN(r=-0.408;p=0.000),HbA1c(r=-0.472;p=0.000)and HOMA-IR(r=-0.590;p=0.000),while positive related to Fincp(r=0.495;p=0.000),ISI(r=0.312;p=0.000),HOMA-β(r=0.391,p=0.000);P was positive related to the expression level to Dkk1 protein(r=0.301,p=0.013).8.Femur was regared as the dependent variable,age,SBP,DBP,BMI,FPG,FMN,TG,TC,LDL-C,HDL-C,Fins,Fincp and other indicators were regared as independent variables,data was analysed by multivariate linear stepwise regression.Fincp was entered into the regression equation in T2 DM group.Multiple linear regression equation was established as follow: y = 1.377-0.339 Fincp.Conclusion: 1.Bone mineral density in postmenopausal women with T2 DM was lower,FPG,FMN,Fincp,25(OH)VitD3 were the influence indicators of it.2.The higher expression level level of Dkk1 protein and gene lead to the lower expression level of β-catenin gene,which may associated with the decrease of bone mineral density in postmenopausal women with T2 DM.
