Objective Filamin A(FLNA)is one of the members in the Filamin family.It was reported that FLNA is involved in a varity of signal transduction in tumors and related to tumor cell proliferation and invasion.However,the relationship between FLNA and pregnancy and preeclampsia is still unknown and rarely explored.This study is aimed to investigate the relationship between the expression of FLNA in placenta during pregnancy,preeclampsia and its clinical indicators of preeclampsia.Methods 1.Subjects: From September 2014 to March 2015,122 pregnant women undergoing maternity and childbirth(or miscarriage)were regularly inspected in obstetrics and gynecology department of the Affiliated Hospital of Qingdao University Medical College.They were divided into early pregnancy group(20 cases),preterm birth group(30 cases),term pregnancy group(32 cases),early-onset preeclampsia group(20 cases)and late-onset preeclampsia group(20 cases).2.The experimental methods: The location and expression levels of FLNA in placenta tissue were detected by the immunohistochemistry,qPCR and western blot.Furthermore,the correlation between placental FLNA expression level and preeclampsia and clinical indicators were analyzed.Result1.FLNA were expressed in placenta tissue of normal pregnancy and preeclampsia group.It is located in cytoplasm,especially on the edge,of villous trophoblast cells,cytotrophoblast cells and vascular endothelial cells;2.The expression level of FLNA mRNA and protein in chorionic villi in early pregnancy group was significantly lower than that in the preterm groupand the full-term pregnancy group(0.32±0.20,0.28±0.12)vs(1.33±0.19,1.13±0.09)and(1.06±0.21,0.98 ± 0.11),(p <0.01);3.The expression level of FLNA mRNA and protein in the placentas in early-onset preeclampsia group was significantly higher than that in the late-onset preeclampsia group and other groups.(1.61 ± 0.22,2.45 ± 0.13)vs(1.38 ± 0.18,1.81 ± 0.14),(p < 0.01);4.The expression level of FLNA mRNA and protein in placenta in early-onset preeclampsia group was significantly higher than that in preterm group,(p <0.01).The expression level of FLNA mRNA and protein in placenta in late-onset preeclampsia group was significantly higher than that in term pregnancy group,(p <0.01);5.The expression level of FLNA in placenta of preeclampsia was significantly correlated with uric acid and urinary protein in 24 hours(r = 0.412 and 0.435,respectively,p <0.01)Conclusions 1.FLNA is involved in the pathophysiology of preeclampsia.2.FLNA expression levels in placental tissue is correlated with the severity of preeclampsia.
