Objective:1.To study the number and phenotype function of decidual macrophages that express CXCR6+/-in the early normal and miscarriage pregnancy;2.To evaluate the functional features of CXCR6+/-decidual macrophages in the early pregnancy.Methods:1.To collect decidual tissues from clinically normal first trimester pregnancies?40cases?and miscarriages?20 cases?and isolate DICs by the method of Percoll Density gradient Centrifugal.2.Decidual macrophages were isolated according to the cell surface markers CD14and by flow cytometry from DICs.3.To study the expression of CXCR6+/-on decidual macrophages by flow cytometry?FCM?.4.To contrast expression levels of M1 surface marker CD80 and M2 surface maker CD206 between the two groups by FCM.5.As well as pro-inflammatory cytokines interleukin TNF-a and anti-inflammatory IL-4?IL-10 were tested by FCM.Results:1.We isolated DICs and the CD14+decidual monocyte-macrophages accounted for approximately 10%of DICS in early pregnancy,and there was no significantly difference between the two groups?P>0.05?.2.The results showed that CXCR6 positively expressed on decidual macrophages,macrophages express CXCR6+in miscarriage pregnancy,significantly lower than the other group?P<0.01?.3.dM?from miscarriage patients express lower level of CD206 and CXCR6+dM?expression higher levels of CD206 than CXCR6-dM?in two groups;dM?in miscarriage groups express higher CD80 and CXCR6-dM?show significantly higher expression of CD80 in miscarriage groups than normal groups.4.Compared to miscarriage groups,normal groups express higher levels of IL-10,there is no significance between the two groups about the expression of IL-4?TNF-a,CXCR6+dM?significantly express higher levels of IL-4?IL-10 than CXCR6-dM?in two groups;Difference from normal pregnancy,CXCR6-dM?significantly express higher levels of TNF-a than CXCR6+dM?in miscarriage pregnancy.Conclusion:Decidual CXCR6+monocyte-macrophages display unique phenotype and function,maybe play a role in miscarriage pregnancy.
