Objective:To investigate the roles of the Th22cells and IL-22in humanfetal-maternal interface during the first-trimester pregnancy. Methods:fifteen women with normal early pregnancy were analyzed byflow cytometry for the percentage of Th22cells in the peripheral blood anddecidua, with thirteen healthy fertile women as control; Meanwhile,enzyme-linked immuno sorbent assay(ELISA) was used to analyze the level ofIL-22in the peripheral blood and deciduawe either.Results:1. The percentage of Th22cells in the peripheral blood of thefirst-trimester pregnancy appeared significantly higher compared to that ofhealthy non-pregnancy(1.11%±0.24%VS0.94%±0.13%)(P <0.05).2. The percentage of Th22cells in the decidua of the first-trimesterpregnancy appeared significantly higher compared to that of the peripheralblood at the same gestation phase(1.33%±0.32%VS1.11%±0.24%)(P <0.05)and that of the peripheral blood at the heathy non-pregnancy women(1.33%±0.32%VS0.94%±0.13%)(P <0.05).3. The levels of IL-22in the peripheral blood of the normal early pregnantwomen were significantly higher than that of the healthy non-pregnant fertile women (104.28±4.73pg/ml vs84.47±5.99pg/ml)(P<0.05). Compared with thelevels of the IL-22in the endometrium of healthy non-pregnant fertile women, that in the decidua of the normal early pregnancy increased significantly(110.47±6.43pg/ml vs88.15±6.49pg/ml)(P<0.05). The levels of IL-22in thedecidua of the normal early pregnancy increased significantly compared to thatin the peripheral blood at the same gestation phase(P <0.05).4. During the early pregnancy, the expression of Th22and IL-22in theperipheral blood was positively correlated statistically(r=0.75, P <0.05). theexpression of Th22and IL-22in the decidua was positively correlatedstatistically(r=0.93, P <0.05).Conclusion:It is speculated that the elevated IL-22may be involved inestablishing and maintaining fetal-maternal immune-tolerance during thefirst-trimester pregnancy....
