Objective:Normal liver function and complete intestinal barrier can maintain the stability of the internal environment. And when the intestinal mucosal barrier is destroyed, endotoxin increases and damages the liver. And when the liver injury develops to a certain extent,it can also counteracts the intestinal mucosal barrier. To investigate the relationship between the changes of intestinal flora in patients with alcoholic liver disease and the liver function(ALT?AST?TBil),and to explore new ideas for the treatment of alcoholic liver disease.Methods:1.A sample of 42 patients with alcoholic liver disease were selected, and they were analyzed in their Escherichia coli,Bacillus coli,Bacillus,Lactobacillus,Bacteroides and Clostridium leptum in the intestinal flora by using Quantitative Real-time Polymerase Chain Reaction Technology.22 cases of healthy persons were selected as control.2.After excluding the patients without liver function serum samples,31 of them were successfully retained in the blood samples, and 20 in the control group. The levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and total bilirubin(TBil)were measured by automatic biochemical analyzer in the sample.SPSS was used for Pearson correlation analysis of the number of bacterial strains in intestinal flora and liver enzyme.Results:1.The Bifidobacterium(5.52±1.73 copies/g)and the Bacteroidetes(7.01±0.72 copies/g)in the feces of ALD patients was significantly lower than that in the healthy control group(P<0.05).There was no significant difference between E.coli,Enterococcus,Lactobacillus and Clostridium leptum in comparison with the healthy group(P>0.05).2.The levels of ALT,AST and TBil in ALD patients were significantly higher than those in the control group(43.47 ± 23.57U/L,40.07 ± 26.55U/L and 17.47 ± 10.71?mol/L,respectively,P<0.05).The correlation analysis of Bifidobacterium and liver biochemical indexes such as ALT,AST and Tbil found that there was a negative correlation between Bifidobacterium and ALT(r =-0.64),AST(r =-0.45)(P<0.01),and negative correlation with TBil(r =-0.11,P <0.05).Conclusion:1.The quantitative analysis of intestinal flora can be carried out by Quantitative Real-time PCR.It is proved that there is intestinal flora imbalance in patients with alcoholic liver disease, and Bifidobacteria and Bacteriobacteria are significantly less than those of healthy people.2.The ALT activity,AST activity and TBil content in the liver of alcoholic patients were significantly increased, and there was a negative correlation between Bifidobacterium and liver functional enzymology index(ALT,AST).
