| Objective:In recent years,the incidence of colorectal cancer has been increasing.It is reported that patients with colorectal cancer have a higher level of intestinal flora imbalance and intestinal inflammation.And probiotics can compete with disease-causing organisms or microbes,resulting in lower concentrations of pathogens or carcinogens in the gut.In addition,hepatocyte growth factor(HGF)and hepatocyte growth factor receptor(c-Met)high expression and amplification can occur in a variety of malignant tumors,such as colorectal cancer,gastric cancer,liver cancer,prostate cancer,ovarian cancer,etc.,And the abnormal activation of c-met can lead to tumor proliferation,invasion,angiogenesis and inhibition of apoptosis.However,it is not clear whether there is a correlation between its specific mechanism and its correlation.The purpose of this study was to investigate the effects of intestinal flora in colorectal cancer on the expression of c-met protein and the level of intestinal inflammation.Furthermore,it was further discussed whether the intestinal flora could affect the invasion and metastasis of colorectal cancer by regulating the HGF/ c-Met signaling pathway.Methods:45 BALB/c mice(6-8 weeks of age,male)were randomly divided into three groups,each group of 15,for antibiotics group,probiotics group and control group,all three groups of mice were given intraperitoneal injection of azomethane(AOM,10.0 mg/kg),Then,four cycles of sodium sulfuric acid glucan sodium salt solution(DSS,2%)and antibiotic solution,probiotics solution and aseptic water were free to drink.(Cycle 1:2 %DSS free drinking dextran sodium sulfate solution 1 week,then antibiotics group was given solution free drink 2 weeks,probiotic group was given probiotic solution free drink 2 weeks,the control group to give free sterile water drinking 2 weeks,respectively,at the same time give each group of antibiotics,probiotics solution and physiological saline solution to fill the stomach,this cycle four times),Anatomical observation on three groups of mice after 12 weeks,collect the intestinal feces,colorectal cancer,colorectal tissues,The intestinal flora analysis was carried out with the second generation of high-throughput sequencing technology in mouse excrement.The colorectal tumor tissue was observed,counted and measured with the naked eye.Then,c-met protein and m RNA were detected by Western Blot and real-time quantitative polymerase chain reaction(q RT-PCR).Pathologic diagnosis was performed by hematoxylin(HE)and pathological methods.Colorectal tissue was used to detect inflammatory factors(IL-1,IL-6,IL-8)by enzyme-linked immunosorbent assay(ELISA).Then the test data are analyzed to determine the difference and correlation of the results.Results:Compared with the control group,the antibiotic group had intestinal flora imbalance.Compared with the control group,the probiotics group significantly improved,and there were a significant number of probiotics.There was no significant difference in the number of colorectal cancer,tumor rate,tumor load and pathological results in the antibiotics group and probiotics group compared with the control group.Moreover,there was no significant difference in c-met expression level between the antibiotic group and the probiotics group compared to the control group.In the antibiotic group,IL-1(P= 0.041),IL-6(P=0.0153)and IL-8(P=0.0006)were significantly higher than those in the control group.And in the probiotics group,IL-1(P=0.0043),IL-6(P=0.0002)and IL-8(P<0.0001)were significantly lower than those in the control group.Conclusions:In colorectal cancer mice,the intestinal flora was correlated with the level of inflammatory factors,and no correlation was found with the HGF/ c-Met signaling pathway. |
