Fatigue is a kind of physiological or psychological phenomenon after the body suffered from long time or high intensive exercise,which shows tired and low productivity.The pathological damage will be occurred when the body is often in a condition of fatigue.It is an effective method to solve the fatigue through giving exogenous supplements or drugs to relieve the occurance of fatigue.Immunity is the ability which can be used to defense disease by the body and maintain the normal physical function.Proper immunity is crucial to the health.Cordyceps have the function of immune regulation,which can enhance the body's resistance to diseases.Therefore,it has a great practical significance to find and screen the active components from natural products and to explore their mechanism of action.Cordyceps fungus is a kind of insect pathogens which belongs to medicinal fungi and has high pharmaceutical value.Ophiocordyceps lanpingensis is a new species of cordyceps,which was found in recent years and belongs to the same genus of Ophiocordyceps sinensis;they are close relatives.The chemical composition of Ophiocordyceps lanpingensis is similar to that of Ophiocordyceps sinensis,and the cultivation of S.is easier than Ophiocordyceps sinensis.It is widely known that Ophiocordyceps sinensis has the functions of enhancing immunity,antioxidation,antivirus and anti-ageing.This study will investigate whether Ophiocordyceps lanpingensis has the effections of antifatigue and enhancing the immunity,thus to confirm the potential of being the alternative of Ophiocordyceps sinensis.This paper studied the effections of antifatigue and enhacing immunity in mice through the motion experiment and various biochemical indicators.Such results provided theoretical basis for the application and development of functional food.The research was carried out from the three aspects below.1.Detection of heavy metal content in artificial O.lanpingensis powder: the samples were processed by wet digestion method,and heavy metal detection was performed using ICP-OES.Results: O.lanpingensis powder contains trace amounts of aheavy metal elements Results: Cadmium ? 0.05 mg/kg,Chromium ? 0.5 mg/kg,16.1? Manganese ? 25.90 mg/kg,34.4? Iron ? 43.0 mg/kg,2.95? Copper ? 4.50 mg/kg,121? Zinc ? 273 mg/kg,0.25? Nickel ? 0.34 mg/kg,0.05? Lead ?0.16 mg/kg,0.16? Arsenic ? 0.23 mg/kg,0.01? Hg ? 0.03 mg/kg.Conclusion: O.lanpingensis powder contains trace amounts of heavy metals,but its content is much lower than the national standard.The establishment of a fast and feasible heavy metal detection system is an important step to achieve its safe,effective,stable and controllable goal.2.The effect of artificial O.lanpingensis extract on anti-fatigue of mice: mice were randomly divided into 4 group,each experiment set is divided into five dose group,respectively low dose group(450 mg/kg),middle dose group(900 mg/kg),the high dose group(1800 mg/kg),and blank control group(distilled water),positive control group(American ginseng 600 mg/kg),continuous lavage for 30 days.The changes of related indexes in mice were measured with the weight swimming experiment of mice,and the indexes of blood lactic acid,serum urea nitrogen,liver glycogen,antioxidant enzyme and blood related parameter were determined.Results: O.lanpingensis extract can increase the swimming time of the mice(low,medium and high dose p<0.05).The concentration of urea nitrogen(p<0.05)was reduced in the middle and high dose group after exercise.The high dose group and the western ginseng dose group could increase the reserve amount of liver glycogen in mice(p<0.05).The decrease area of 3 time point curve of lactic acid was lower than that of blank control group,but there was no significant difference between groups,at the same times.O.lanpingensis extract can raise the blood hemoglobin and blood sugar content.It can significantly increase the activity of antioxidant enzymes and reduce the blood malondialdehyde concentration.Conclusion: the study shows that the mycelium of O.lanpingensis extract has the effect of relieving fatigue.3.The effect of artificial O.lanpingensis extract on immune deficiency of mice: the immune deficiency model was constructed with cyclophosphamide immuno suppressive agents.This experiment through the separation of the spleen in vitro,obtain the original generation of mice spleen cells,T cell activation by Con A induced,This study will investigate whether O.lanpingensis extract has the effect on proliferation activity of mice spleen T lymphocytes,T cell cycle,cytokine expression level,inspection indexes of the clearance rate of carbon,antibody generation cells(modified glass Jeren method),the mice immune factor IL-2,IL-6,TNF-?,IL-10.Delayed-type hypersensitivity mice model was established,the analysis of O.lanpingensis extract of delayed-type hypersensitivity in mice induced by DNFB pharmacological effects.Results: low,medium and high three dose groups of mice were given after 30 d,O.lanpingensis extract increased the immune organ index in mice,O.lanpingensis extract(high dose groups)can improve the late onset of allergy of mice,and promote the mice antibody generation cell proliferation,stimulating the secretion and expression of cytokines,it has the function of improving celluar immunity and humoral immunity.It can be determined that O.lanpingensis extract can enhance the immunity of mice.
