BackgroundAdipsin,which belongs to serine proteinase,was the first adiponkine described.In the alternative pathway of complement activation,adipsin catalyzes the rate-limiting step and the production of C3adesArg(ASP),and were so called complement factor D(CFD).It has been found that adipsin involves in the pathology of the ischemia-reperfusion injury and sepsis,however,little were known about its relationship with metabolic disorders.Recently,Lo et al found that mice lacked adipsin became glucose intolerance.The fasting plasma glucose of db/db mice after received adipsin were significantly decreased,also,the glucose deposition rate and glucose stimulated insulin secretion were remarkably increased on fasting condition.Zhang et al found that the glucolipid metabolism and insulin resistance of ZDF type 2 diabetes mellitus mice were greatly improved after received adipsin.Nevertheless,the relationship between serum adipsin and pancreatic ?cell function and insulin resistance in human being has not been explored,and the mechanism are still unclear.ObjectiveTo detect the levels of serum adipsin in different glucose tolerant people,and investigate the relationship between serum adispin and pancreatic ? cell function.MethodsFifty-six patients with newly diagnosed type 2 diabetes mellitus(T2DM),36 patients with newly diagnosed impaired glucose regulation(IGR),and 45 subjects with normal glucose tolerance(NGT).All participants underwent intravenous glucose tolerance tests.Acute insulin reaction(AIR)and 0-10 min the area under the insulin curves(AUC)were calculated.The homeostasis model assessment of pancreatic ? cell function index(HOMA-?)and insulin resistance index(HOMA-IR)were calculated.The serum adipsin and IL-? were assayed by ELISA.ResultsThe serum adispin levels were significantly lower in IGR and T2DM(P<0.05);correlation analysis showed that serum adispin levels were negatively correlated with WHR,FFA,FPG,2hPG,HbAlC,HOMA-IR,IL-1? and hs-CRP(P<0.05 or P<0.001),positively correlated with HOMA-?,HDL-C,AUC,and AIR inversely(P<0.05 or P<0.001);stepwise regression analysis revealed that HOMA-? and AIR were independently related with serum adispin.ConclusionSerum adispin levels were significantly decreased in IGR and T2DM,and were closely related with glucolipid metabolism and the first-phase of insulin secretion.Adipsin might involve in the development of T2DM.
