Objective:To study the effect of Shi Wei Jiang Tang San on the antioxidant capacity of glucose lipid metabolism and liver in diabetic rats,and to provide theoretical basis for its development and utilization;To observe the effect of Shi Wei Jiang Tang San on the expression of hepatic and fat AMPK,and to find possible mechanism for the improvement of glucose lipid metabolism.Methods:The four-week old male SD rats were bred for 1 week,feeding high-sugar and high-fat feed for 4 weeks,and then injected STZ(45mg/kg)in the abdominal cavity.After 72 hours of injection,the fasting blood glucose of rats was greater than 16.7mmoL/L.The rats with stable model were randomly divided into the model group and the Shi Wei Jiang Tang San group,10 rats in each group.The model group and the ten-taste hypoglycemic group were fed with the same amount of normal saline and 10 flavorous drops(0.003mL/g),respectively,and fed the high-sugar and high-fat feed.In addition,10 rats were randomly selected for the blank group and fed normal feed without any treatment.Observe the water intake,urine volume and mental state of rats daily.Weekly test between groups of rats after fasting blood sugar and weight,in 2,4,6,8 weeks orbital blood collection,preparation,serum and kit to measure each group rats serum superoxide dismutase(SOD),malondialdehyde(MDA).In the 8th week,the blood was weighed and executed,and the same side of the epididynia was weighed and the visceral fat content was calculated,and the liver was located in the same position.With the kits in the rat liver glycogen content and serum cholesterol(TC),triglyceride(TG),high-density lipoprotein(HDL),low density lipoprotein(LDL)and the content of adiponectin and measured glycosylated hemoglobin levels.(2)the expression and quantitative analysis of liver and fat AMPK in rats were detected by immunoblotting.Results:(1)the fasting blood glucose of the rats in the model group and the Shi Wei Jiang Tang San group was higher than that in the blank group(p < 0.01).Compared with the model group,the blood glucose showed no statistical significance(p BBB 0.05).The glycosylated hemoglobin was lower than that in the model group,but there was no statistical significance(p BBB 0.05).The content of hepatic glycogen was significantly higher than that in the model group(p < 0.01).(2)compared with the blank group,2,4,6,8 weeks of model group and 10 flavour hypoglycemic SOD activity decreased significantly(p < 0.05),6,8 weeks ten taste the SOD activity of hypoglycemic scattered group was obviously higher than that of model group(p < 0.05);2,4,6,8 weeks,model group and 10 flavour hypoglycemic MDA content is higher than that of blank group(p < 0.05),MDA content and 10 flavour hypoglycemic scattered group were less than model group,of which 6,8 weeks,significant difference(p < 0.05).(3)model group and 10 flavour hypoglycemic serum TC,TG,LDL scattered group were significantly higher in the blank group(p < 0.05),HDL decreased significantly(p < 0.05),10 flavour hypoglycemic scattered groups of serum TC,TG,LDL,compared with model group were lower(p < 0.05),HDL rose(p < 0.05);Model group and 10 flavour hypoglycemic serum adiponectin levels lower than the blank group(p < 0.05),10 flavour hypoglycemic than model group serum adiponectin level is reduced,but no statistical difference(p > 0.05);The visceral fat content was significantly increased(p < 0.01),and the visceral fat was lower than that in the model group(p < 0.05).(4)compared with the blank group,the level of the hepatic AMPK in the group of 10 taste hypoglycemic group and the model group decreased significantly(p < 0.01),and the level of AMPK in the Shi Wei Jiang Tang San group was lower than that in the model group(p < 0.05).There was a slight difference in the level of fat AMPK in the Shi Wei Jiang Tang San group and the model group,but there was no statistical significance(p > 0.05).Conclusion :(1)Shi Wei Jiang Tang San can reduce the content of glycosylated hemoglobin,increase the content of glycogen,and regulate the disorder of glucose metabolism caused by diabetes.(2)Shi Wei Jiang Tang San can enhance the activity of serum SOD,reduce the content of MDA and improve the antioxidant capacity of diabetic rats.(3)Shi Wei Jiang Tang San can reduce serum TC,TG,LDL,adiponectin content,increase serum HDL content,reduce visceral fat content,and improve the body's fat metabolism disorder.(4)Shi Wei Jiang Tang San can increase the expression of AMPK? in the liver,but it has no effect on the expression of fat AMPK?.The improvement of glucose and lipid metabolism may be related to the liver AMPK?.
