| ObjectiveThe project aims to establish a simple high-fat diet induced IGT r-at model.Which is adopted to observe the effects of Huayuwendan Tang on the appearance change and the pathological characteristics.It is to present the mechanism of Huayuwendan Tang to improve IR and the m-echanism of energy metabolism,and to support the medical target with experimental data.MethodsMale healthy Wistar rats were randomly divided into two groups.T-he rats in the normal group were fed with normal diet,while the other group of rats were fed with high fat diet,for 20 weeks.Then they were obseved and measured from appearance as well as IPGTT,FPG and 2hP G.The successful model rats were randomly divided into model group,TCM group,Western Medicine group.The TCM group was given Huayuw-endan Tang by 5.2g/kg d,The Western Medicine group was given metfor-minsolution by 99mg/kg d,the control group and model group were give-n the same volume of saline.After continuous administration for 4 week s.At the end of the experiment,the experimental subjects were conducted,the abdominal aorta was collected,the serum was separated.Fins,GSP,AD PN and LEP were detected by ELISA,and IAI and IRI were calculated.HDL-C and LDL-C were detected by biochemical analyzer.The liver qu-ality was weighed up and liver index was calculated.Liver homogenate was used to detect TG,TC,LPO.Glycogen storage was observed by liver glycogen staining.The target density and average gray level were analy-zed by image analysis software.The expression of AMPKa and ACCa mRNA in the liver AMPK signaling pathway was detected by Realtime PCR.Western blot was used to detect the expression of AMPKα,p-AMP Ka,ACCa,p-ACCa protein.Research results1.Index of serum glucose,serum lipid,insulin sensitivity:Huayuwend-an Tang reduced weight,and the FPG and 2hPG of rats,which were stat-istically significant compared with the model group(p<0.05 or p<0.01),and there is timeliness when it reduced the FPG.Huayuwendan Tang de-creased Fins,IRI,GSP levels,while increased IAI levels,which were statis-tically significant(p<0.05)compared with the model group.Huayuwendan Tang improved serum HDL-C levels,while reduced LDL-C levels,which were statistically significant(p<0.01)compared with the model group.2.Cytokines:Huayuwendan Tang improved serum ADPN levels,while reduced LEP levels,which were statistically significant(p<0.01)compared with the model group.3.Liver glucose and lipid metabolism,oxidative stress index:Huayuw-endan Tang reduced liver quality,liver index,TG and TC,which were sta-tistically significant compared with the model group(p<0.05 or p<0.01).Huayuwendan Tang reduced the level of LPO and increased the level of GSH-Px,which were statistically significant compared with the model group(p<0.05 or p<0.01).Huayuwendan Tang increased the target densit-y and average gray level,which were significantly different from that of model group(p<0.01),by increasing liver glycogen reservior.4.Hepatic AMPK signal pathway:Huayuwendan Tang improved the expression of AMPKa mRNA in liver tissue,which was statistically sign-ificant compared with the model group(p<0.05).And it had no effect on the expression of ACCa mRNA(p>0.05).Huayuwendan Tang improved the expression of AMPKa,p-AMPKa and p-ACCa protein in liver tiss-ue,which were statistically significance compared with the model group(p<0.05 or p<0.01).And it had no significant effect on the expression of ACCa protein(p>0.05).Conclusion1.Huayuwendan Tang has a therapeutic effect on the model rats,it can reduce blood glucose and blood lipid levels,correct the metabolism disorders of glucose and lipid,and improve IR.2.Huayuwendan Tang can increase the level of ADPN,reduce the le-vel of LEP and improve the energy metabolism in model rats.3.Huayuwendan Tang can reduce the liver fat deposition,improve t-he oxidative stress state,increase the glycogen storage in liver tissue of the model rats,and play a role in protecting the liver.4.Huayuwendan Tang can improve the liver AMPK activity,inhibit ACC activity of the model rats,inhibit the synthesis of fatty acids,incre-ase the oxidation of fatty acids,improve the energy metabolism,and incr-ease insulin sensitivity.5.Huayuwendan Tang can regulate the AMPK-ACC signaling pathway.The effect of Huayuwendan Tang on hepatic AMPK-ACC signaling path-way is one of the targets to improve IGT IR. |
