Objective:By establishing a sports fatigue mouse model and an electrically stimulating specimen from the gastrocnemius muscle of toad,the alleviating effect of Lycium ruthenium decoction on sports fatigue was explored,and the related mechanism was discussed.Method:1.200 mice were divided into five groups,namely:Lycium ruthenicumM.low?3mg/g?,medium?6mg/g?and high dose groups?12 mg/g?,control group(0.2ml?10g-1)and blank group?0.2ml/10g?,40 rats/group.Animals of each group were again randomly assigned for 4 groups,10 rats in each group:exhaustive exercise group,no swimming group,immediate after swimming group and restore 30min after swimming group.Continuous administration of 30d,the fatigue model of mice was established by swimming for 40min in the?25±2??water.The time of exhaustive swimming was determined.The differences of blood glucose,muscle glycogen and liver glycogen,serum urea nitrogen and lactic acid were measured before,afterand30min after swimming.2.20 specimens of gastrocnemius from toad were prepared,and 2 gastrocnemius specimens of the same toad were randomly divided into control group?Ringer's solution group?and observation group?Lycium ruthenicum Decoction group?.The gastrocnemius muscle was stimulated by pulsed current,and the effects of drugs on the amplitude of muscle contraction,the time required to reach maximum contraction amplitude and the duration of continuous contraction of muscle were observed.The mechanism of calcium free medium and calcium channel blocker was discussed.Results:1.Compared with blank group,the high and medium doses groups of Lycium ruthenicumnot only significantly prolonged exhaustive swimming time?P<0.05?,but also increased liver glycogen and muscle glycogen reserve,reduced serum urea nitrogen level and reduced lactic acid production?P<0.05?.2.Lycium ruthenicum decoction can shorten the maximum time required for muscle contraction,prolong T50%and T10%,and shorten the time required for fatigue recovery.After calcium channel blockers added,the effect was decreased obviously.Conclusion:1.Lycium ruthenicumM.could relieve the fatigue in exercise mice.2.Lycium ruthenicum decoction could enhance the contractility of toad gastrocnemius in vitro,and alleviate the contraction fatigue induced by electrical stimulation.The effect may be related to the promotion of calcium influx and the increase of intracellular calcium concentration.
