ObjectiveIn this research,we deteced the expression of IL-28 B in the peripheral blood,which contained CIN? group,CIN?I-CIN? group,cervical cancer(preoperative)group,cervical cancer(postoperative)group and the control group.We discussed the genotype and gene frequency of the single nucleotide polymorphisms(SNP)sites of IL-28B(rs12979860 and rs8099917)and try to analyze the correlation between them and the incidence of cervical cancer.And we analyzed the reason of expression differences and explored its role in the development of cervical cancer.MethodsData and anticoagulant blood of 10 cases for CIN? group,20 cases for CIN?I-CIN?group,30 cases for each group of cervical cancer(preoperative)group,cervical cancer(postoperative)group,and the control group were all collected.IL-28 B single nucleotide polymorphisms(SNP)of rs12979860,rs8099917 were tested by PCR and gene sequencing to analysis the correlation between the genotype and allele frequency of IL-28 B and incidence of cervical cancer.The expression of IL-28 B mRNA in peripheral blood were examined by RT-PCR.Result:1.At the rs12979860 site of IL-28 B,among the 10 patients in CIN? group,the genotype of C/C was 8(80.00%),C/T was 2(20.00%),and the frequency of C allele was 90.00%,and the frequency of T allele was 10.00%.Among the 20 patients in CIN?I-CIN?,the genotype of C/C was 16(80.00%),C/T was 4(20.00%),and the frequency of C allele was 90.00%,and the frequency of T allele was10.00%.Among the 30 patients with new cervical cancer,the C/C genotype was 25(88.33%),C/T was 5(16.67%),and the frequency of C allele was 91.67%,and the frequency of T allele was 8.33%.Among the 30 patients with cervical cancer treatment,the C/C genotype was 26(86.67%),C/T was 4(13.33%),and the frequency of C allele was93.33%,and the frequency of T allele was 6.67%.Among the 30 healthy control groups,the C/C genotype was 28(93.33%),C/T was 2(6.67%),and the frequency of C allele was 96.67%,and the frequency of T allele was 3.33%.The rs12979860 site of IL-28 B had no statisticallysignificant difference in the frequency of genotypes and alleles in each group(P=0.66,P=0.71).2.The rs8099917 site of IL-28 B,among the 10 patients in CIN? group,the genotype of T/T was 8(80.00%),T/G was 2(20.00%),and the frequency of T allele was 90.00%,and the frequency of G allele was 10.00%.Among the 20 patients in CIN?I-CIN? group,the genotype of T/T was16(80.0%),T/G genotype 4(20.0%),the frequency of T allele was 90.0%,and the frequency of G allele was 10.0%.Among the 30 patients of new cervical cancer the T/T genotype 23(76.67%),T/G genotype 7(23.33%),and the frequency of T allele was 88.3%,and the frequency of G allele was 11.67%.Among the 30 patients with cervical cancer treatment,the T/T genotype 25(84.6%),T/G genotype 5(17.5%),and the frequency of T allele was91.67%,and the frequency of G allele was 8.33%.Among the 30 healthy control groups,the T/T genotype 26(86.7%),T/G genotype 4(13.3%),and the frequency of T allele was93.33%,and the frequency of G allele was 6.67%.The rs8099917 site of IL-28 B had no statistically significant difference in the frequency of genotypes and alleles in each group(P=0.86,P=0.88).3.The expression of IL-28 B mRNA in peripheral blood was as follows:CIN? group(99.23±11.67),CIN?I-CIN? group(78.87±21.97),cervical cancer(preoperative)group(20.90±8.89),cervical cancer(postoperative)group(192.34±22.65)and the control group(308.56±34.55).The expression of IL-28 B in each group was lower than that in the control group(P<0.05).Each group of IL-28 B expression gradually decreased with lesion degree aggravating,namely the expression of IL-28 B is stronger in CIN? group than that in CIN?I-CIN ? group(P<0.05),the expression of IL-28 B is stronger in CIN ?I-CIN ? group than that in cervical cancer(preoperative)group(P<0.05).The expression of IL-28 B was different before and after surgical treatment,that is,he expression of IL-28 B in cervical cancer(postoperative)was stronger than that of cervical cancer(preoperative),and the difference was statistically significant(P<0.05).4.In cervical cancer(preoperative)and cervical cancer(postoperative)group the expression of IL-28 B in lymphatic metastasis negative group was stronger than that in lymphatic metastasis positive group,and the difference was statistically significant(P<0.05).Conclusion:1.In our research,compared with IL-28 B wild type rs12979860 CC type and rs8099917 TT genotype,mutation site genotype(TC,TG)did not increase the risk of cervical disease.2.There was no significant correlation between IL-28 B rs12979860,rs8099917 site genotype,gene frequency and cervical cancer incidence.3.The expression of IL-28 B may play an important role in the development and metastasis of cervical cancer.
