Objective: MCI is a common disease in the elderly,which is characterized by a further decline in cognitive function with age,which is usually considered as an early warning signal of AD.We believe that effective intervention for MCI is an effective measure to delay or even reverse the progress of AD.Traditional Chinese medicine(TCM)believes that the constitution type of MCI is mainly composed of Yang deficiency,Qi deficiency and blood stasis,so it can be used to treat MCI by Strengthening Qi and warming yang and promoting blood circulation.Dunhuang weicifang also known as the four party service often,written in accordance with the ancient books,the hidden in the ancient medical scientist Wei Ci,so the name of Dunhuang weicifang.Dunhuang four party service often comes from the Dunhuang suicide note,the syndrome of insufficiency of kidney yang deficiency and Yang is,go up,make fire hyperactivity in,and mean to kidney failure,cause insomnia,waist weakness,distrait;dizziness,tinnitus and deafness,aversion to cold is kidney yang deficiency,Qi deficiency syndrome;its main treatment method is Wenshen impotence,jiaotongxinshen.In order to clarify the effect of Dunhuang sishi chang fu fang take on learning and memory function of rats,this study through the water maze test and step-down test of different group of learning and memory in rats were detected at the same time,compare on biochemicalindexes;by detecting the cell morphology of each group were detected by cell morphology and apoptosis of brain cells explore the brain tissue of mice dying situation.Western blot was used to detect p35,CDK5 and ERK protein expression.Methods:Chose 70 male cleaning degree rats,and divided them into model groups according to the random number:low dose group,medium dose group,high dose group of sishi changfu fang,Positive drug control group,sham-operated group and blank group,each group contained 10 rats.Inject the first six groups of the hippocampal CA1 region by condensed state A?1-42,each 5?l and established the rat AD model.After establishing the model for 7 days,gave lavage administration for 28 days(100ml/kg per day);gave model group and sham-operated group saline,gave sishi changfu fang group diffrent concentrations and give positive drug control group donepezil.After lavage administration,seperate the hippocampal and stored in-18?.Gave Morris water maze(MWM)and step-down test on rat's learning and memory abilities;at the same time examined the rats' bio-chemical index;took Light microscope slices to do research on brain tissue cell morphology;took flow cytometer method to test the INOS expression quantity in the hippocampal tissue cell;use ELISA to test the condition of the P35?CDK5?ERK protein expression.Results :1 the experimental results showed that the model group and the group of four times in the sham operation group and the normal group had different degrees of escape latency.Especially the escape latency of model group increased the most obviously,the rest of the treatment groups have a certain degree of escape latency shortened,and the high dose of four party service groups often have significant difference(P<0.05);the positive drug control group and four party service often group and model group compared with significant difference(P<0.05),while the experimental results 2 the model group showed that the latency and number of errors was significantly higher than that in other groups,the rats in the model group on the original platform lost memory.The performance of each medication group was significantly higher than that of the model group,compared with the model group(P<0.05).3 flow cytometry was used to measure the expression of INOS in cells.As shown from the flow cytometry to detect the intracellular INOS expression analysis showed that the four party service,often and positive drug can reduce the expression of INOS in cells,so as to achieve the protective effect of different concentrations of four,but often take side comparison,the high dose of four often served soup the effect of low dose group,significant difference(P<0.05).4.Compared with the control group,the content of ERK in the model group was significantly lower than that in the control group,and ERK levels in the treatment group changed to some extent.Compared with the sham operation group,the positive expression of P35 was significantly increased(P<0.05),and the most obvious increase was in the positive drug control group(P<0.05).P35 positive cells in the treatment group decreased to some extent compared with the control group,P35 positive cells in the low dose group were lower than those in the control group(P<0.05),P335 positive cells in the medium dose group and the high dose group were significantly lower than those in the control group(P<0.05).There was no significant difference in P35 positive cells between the treatment group and the control group(P> 0.05).The expression of cdk-5 protein in brain tissue of model group was significantly higher than that of control group and sham operation(P<0.05),and the expression of cdk-5 protein in model group was significantly higher than that of model group(P<0.05).There was no significant difference in protein expression between low,medium and high dose groups(P> 0.05).Conclusion: four frequently used prescription and positive drugs can improve the learning and memory ability of rats in different groups.The high dose four is often used in the lower dose of four is often better in terms of service.The four party service and Donepezil Hydrochloride Tablets often can improve the physiological indexes of mice.The Dunhuang four party service often can
