| Objective: The aim of this study was to isolated exosomes released by toungue cancer cell supernatant and analysis exosomal Micro RNA expression profoile.Methods: Exosomes were isolated from Tscca cells supernatant by kits and inspected by scanning electron microscopy,quantified via nanoparticle tracking analysis,verified by flow cytometry labeled CD9 and CD81,in these means did a preliminary analysis of the morphological and molecular biology of the exosomes.The expression profiles of Tscca cells derived exosomes Micro RNA were studied by high flux sequencing technology,the potential targets of the Micro RNA were predicted.Gene Ontology and Kyoto Eneyclopedia of Genes and Genomes analysis were performed to determine the exosomes Micro RNA.Results: The typical cup-shaped structure of the exosomes was isolated from the Tscca cell supernatant,which was less than 150 nm,and the marker protein CD9 and CD81 were expressed as exosomes.We analyzed micro RNA expression in exosomes from Tscca cells,in total,247 Micro RNAs,bioinformatical analysis revealed that 72 Gene Ontology terms and 46 signaling pathways were associated with the potential targets of exosomesal Micro RNA.Conclusion: Tongue cancer cells secrete exosomes,which are rich in a variety of Micro RNAs related to cell metabolism and cycle,suggesting that they play an important role in the occurrence and evolution of tongue cancer. |
