Effect Of Different Resuscitation Fluids On Interleukin-8 And Tumor Necrosis Factor-α In Rats With Traumatic Hemorrhagic Shock

Objective : To investigate effect of different resuscitation liquids and various resuscitation methods on contents of interleukin-8(IL-8)and tumor necrosis factor-α(TNF-α)in early resuscitation process of rats with traumatic hemorrhagic shock.Methods:Eighty healthy adult clean male SD rats(250~320 g,10 to14 weeks)were chosen and divided into 5 groups randomly and averagely: shock control group,crystal liquid limited resuscitation group,colloidal liquid limited resuscitation group,7.5% Na Cl limited resuscitation group,and colloidal liquid non-limited resuscitation group.There were 16 rats in each group.The model of hemorrhagic shock we adopted was created by Capone et al.Rats in each shock control group and each experimental group were weighed before intraperitoneal injection of pentobarbital sodium anesthesia and insert catheters into left femoral artery and right femoral artery after anesthesia,after the blood pressure is stable for last 10 minutes.Animal model was established via Chaudry’s method,the experimental animals in shock control group didn’t receive any resuscitation treatment,and we recorded the parameters and sacrificed the experimental group rats at the 4 times points.The rats were sacrificed in batches at the time points of 2 hours,6 hours,12 hours and 24 hours after anesthesia.Then the rats were killed,and the experiments were terminal.The contents of IL-8 and TNF-α in serum were detected by enzyme linked immunosorbent assay.Results: 1.The rats in shock control group died successively after shock state were sustained for 1 hour,time ranged from 20 min to 50 min.The content of IL-8 and TNF-α in the serum of these rats are higher than each experimental group consistently.(P<0.05)2.After different treatments in rats with hemorrhagic shock,the contents of IL-8 and TNF-α among three kinds of limited resuscitation groups(crystal liquid limited resuscitation group,colloidal liquid limited resuscitation group,7.5% Na Cl limited resuscitation group)on hour 6 after resuscitation were significantly higher than those on hour 2 after resuscitation(P<0.05)and reached the peaks,then began to decrease.With the extension of time in shock,on hour 12 after resuscitation,the contents of IL-8 andTNF-α were decreased continuously among three kinds of limited resuscitation groups(crystal liquid limited resuscitation group,colloidal liquid limited resuscitation group,7.5% Na Cl limited resuscitation group)(P<0.05).3.The contents of IL-8 and TNF-α in colloidal liquid non-limited resuscitation group at each point time were significantly higher than those among three kinds of limited resuscitation groups(crystal liquid limited resuscitation group,colloidal liquid limited resuscitation group,7.5% Na Cl limited resuscitation group)(P<0.05).4.The contents of IL-8 and TNF-α in crystal liquid resuscitation groups(crystal liquid limited resuscitation group and 7.5% Na Cl limited resuscitation group)were significantly lower than those in the other limited liquid resuscitation groups(P<0.05).Conclusions: 1.In process of liquid resuscitation of rats with traumatic hemorrhagic shock,limited resuscitation method is better than that of non-limited resuscitation method.2.Among three kinds of limited resuscitation methods,crystal resuscitation liquid is more effective than the other two resuscitation liquids(crystal liquid limited resuscitation group and 7.5% Na Cl limited resuscitation group)in prohibiting releases of IL-8 and TNF-α in rats with traumatic hemorrhagic shock.
